Study on the active components of Sanhuang Xiexin Decoction in inhibiting α-amylase and α-glucosidase activities
Objective To screen the effective parts of Sanhuang Xiexin Decoction for inhibiting the inhibitory activities of α-am-ylase and α-glucosidase,and to study its active components.Methods The aqueous decoction of Sanhuang Xiexin Decoction was eluted with macroporous resin to obtain the aqueous part,30%ethanol part,60%ethanol part and 95%ethanol part,and the a-queous decoction and the four eluted parts were subjected to α-amylase and α-glucosidase inhibition tests in vitro to screen the best active parts,and then analyzed the active components by high performance liquid chromatography,and the methodological in-vestigations and content determinations were carried out.Results The best site for inhibiting α-amylase activity was screened to be the 30%ethanol site,with an IC50 of 1.707 mg·mL-1,and the active ingredients were baicalin,berberine,wogonoside,ba-icalein,aloe-emodin,rhein and emodin,with baicalin being the most abundant(177.588 mg·g-1),and wogonoside being the second most abundant(30.546 mg·g-1);The best site for inhibition of α-glucosidase activity was the 60%ethanol site with an IC50 of 0.0123 mg·mL-1,and the active ingredients were baicalin,berberine,wogonoside,baicalein,aloe-emodin,rhein,emodin,chrysophanol,and physcion,with baicalin being the most abundant(181.631 mg·g-1),and berberine being second most abundant(71.724 mg·g-1).Conclusion Sanhuang Xiexin Decoction has active inhibitory effect on both α-amyl-ase and α-glucosidase to a certain extent,and the inhibitory effect on α-glucosidase is better than that on α-amylase,which is of some reference value for the development of α-glucosidase inhibitors.
