补阳还五汤改善糖尿病心肌损伤的配伍作用研究

目的 观察补阳还五汤及其配伍对糖尿病小鼠心肌损伤的保护作用及其可能的作用机制,为临床应用提供理论基础和实验依据。方法 以高脂饲料联合链脲佐菌素制备糖尿病模型小鼠,通过检测血糖、TC、TG、LDH、CK-MB、IL-2、TNF-α、MDA、SOD的水平;HE、Masson染色检测心肌组织病理学变化,Western Blot法检测心肌组织中MFN2、DRP1、MFF蛋白表达水平;观察补阳还五汤及其不同配伍组方干预糖尿病心肌损伤的作用,初步分析其作用机制。结果 与模型组相比,补阳还五汤组的血脂明显降低(P<0。05),血清LDH、CK-MB、IL-2、TNF-α明显下降(P<0。05),补阳还五汤组和活血组心肌SOD活性明显升高(P<0。05),各给药组MDA含量明显降低(P<0。01)。病理染色结果显示,与正常组比较,模型组动物心肌细胞间隙变大排列不均,存在心肌损伤,心肌胶原纤维增多,经各组给药治疗,心肌病理变化有所改善。Western blot检测结果显示,与模型组相比,各组给药上调MFN2蛋白表达,下调DRP1、MFF蛋白表达。结论 补阳还五汤可减轻糖尿病心肌损伤,其作用可能与抑制炎症反应缓解氧化应激损伤及调控线粒体动力学蛋白表达相关。
Study on the compatibility mechanism of Buyang Huanwu Decoction on improving myo-cardial injury in diabetic
Objective To observe the protective effect of Buyang Huanwu Decoction and its compatibility on myocardial injury in diabetic mice and its possible mechanism,so as to provide theoretical basis and experimental basis for clinical application.Meth-ods Diabetic model mice were prepared with high-fat diet combined with streptozotocin,and the levels of blood glucose,TC,TG,LDH,CK-MB,IL-2,TNF-α,MDA and SOD were detected;HE and Masson staining were used to detect the patholog-ical changes of myocardial tissue,Western Blot method was used to detect the protein expression levels of MFN2,DRP1 and MFF in myocardial tissue;To observe the effect of Buyang Huanwu Decoction and its different compatible prescriptions on diabetic myocardial injury,and preliminarily analyze its mechanism.Results Compared with the model group,the blood lipids in the Buy-ang Huanwu Decoction group were significantly lower(P<0.05),and the serum LDH and CK-MB were significantly lower(P<0.01).The results of pathological staining showed that compared with the normal group,the gaps between the myocardial cells in the model group became larger and the arrangement was uneven,there was myocardial damage,and the myocardial collagen fi-bers increased.The results of Western blot showed that,compared with the model group,administration of each group up-regu-lated the expression of MFN2 protein,and down-regulated the expression of DRP1 and MFF protein.Conclusion Buyang Hua-nwu Decoction and its compatibility can reduce myocardial injury in diabetic mice,and its mechanism may be related to inhibiting inflammatory reactin,alleviating oxidatire stress injury,and Regnlating the expression of mitochondrial dynamic proteins.
