萸连丸对DSS诱导结肠炎小鼠Th17/Treg平衡的调控机制

目的 探讨萸连丸对葡聚糖硫酸钠(DSS)诱导的溃疡性结肠炎(UC)疗效和Treg/Th17平衡的调控机制。方法 将BALB/c雄性小鼠随机分为正常组、模型组、萸连丸组和美沙拉嗪给药组,每组10只。除正常组外,其余各组采用"2。5%DSS(第1~7天)——自由饮水(第8~14天)——2。5%DSS(第15~21天)"方法复制慢性复发性结肠炎。同时造模第8天开始予以萸连丸、美沙拉嗪肠溶片连续灌胃14d。造模第22天观察小鼠部分形态指标,采用流式细胞术测定结肠组织中CD4+Foxp3+、CD4+IL17A+细胞水平,采用蛋白印迹法检测结肠组织中检测低氧诱导因子(HIF-1a)、驱动蛋白家族成员2A(KIF2A)、BCL-2同源结构域蛋白NOXA、蛋白磷酸酶2a(PP2A)表达水平。结果 与模型组相比,萸连丸组小鼠体质量明显增加,结肠长度明显增加,肠重指数明显降低;病理提示结肠组织中少量炎性细胞浸润,溃疡基本修复;结肠组织中CD4+IL17A+细胞水平显著下降,CD4+Foxp3+细胞水平显著升高;结肠组织中HIF-1a、PP2A表达水平显著降低,KIF2A表达水平显著升高。结论 萸连丸可通过Th17/Treg细胞平衡而有效治疗DSS诱导小鼠慢性复发性结肠炎,其作用机制涉及HIF-1a、PP2A表达水平降低和KIF2A表达水平升高。
Regulation of Yulian Pill on Th17/Treg Balance in DDS Induced Colitis Mice
Objective To investigate the therapeutic effect of Yulian Pill(YLP)on dextran sodium sulfate(DSS)-induced ul-cerative colitis(UC)and the regulation mechanism of Treg/Th17 balance.Methods BALB/c male mice were randomly divided into normal group,model group,YLP group and mesalazine administration group,10 in each group.Except for the normal group,the rest of the groups adopted the method of"2.5%DSS(1st to 7th day)-free drinking water(8th to 14th day)-2.5%DSS(15th to 21st day)"to replicate the chronic recurrent colon inflammation.At the same time,YLP and Mesalazine Enteric-coated Tablets were given by intragastric administration for 14 days from the 8th day of modeling.On the 22nd day after modeling,some morphological indicators of the mice were observed,the levels of CD4+Foxp3+and CD4+IL17A+cells in the colon tissue were measured by flow cytometry,and the levels of hypoxia-inducible factor(HIF-1α),Kinesin family member 2A(KIF2A),(NOXA),protein phosphatase 2a(PP2A)expression levels.Results Compared with the model group,the body weight of the mice in the YLP group was significantly increased,the length of the colon was significantly increased,and the intestinal mass in-dex was significantly decreased;pathology showed that a small amount of inflammatory cells infiltrated the colon tissue,and the ulcer was basically repaired;The level of CD4+IL17A+cells in the colon tissue decreased significantly,and the level of CD4+Foxp3+cells increased significantly;the expression levels of HIF-1α and PP2A in the colon tissue decreased significantly,and the expression level of KIF2A increased significantly.Conclusion YLP can effectively ameliorated DSS-induced chronic recur-rent colitis in mice by regulating the balance of Th17/Treg cells,and its mechanism of action involves the decreased expression levels of HIF-1α,PP2A and the increased expression level of KIF2A.
