Genetic diversity analysis of Cultivated Species Dendrobium officinale in Guizhou
Objective To study the genetic diversity of Dendrobium cultivars in Guizhou,which can provide a reference for germ-plasm resource utilization and genetic improvement.Methods Using Inter-simple sequence repeats(ISSR)and Simple se-quence repeat(SSR)molecular marker techniques,we analyzed the genetic diversity of 20 cultivars of Dendrobium from Zhejiang and Yunnan in Guizhou,and analyzed the results using NTsys and Popgen32 software.Popgen32 software was used to analyze and study the results.Results Dendrobium ironbark was subjected to DNA molecular markers,and 32 primers amplified 152 bands with an average of 4.75 bands per primer,including 145 polymorphic bands with an average of 4.53 polymorphic bands per prim-er,and the polymorphism rate(PPL)was 96.05%.The average observed allele number(Na)of the 20 Dendrobium samples was 1.9605,the average effective allele number(Ne)was 1.5992,the average Nei's gene diversity index(H)was 0.3507,and the average Shannon's polymorphism information index(I)was 0.5219 by Popgen32 analysis.The genetic variation was abun-dant.The clustering analysis by UPGMA showed that the 20 Dendrobium samples could be clustered into two classes,indicating that the Zhejiang and Yunnan Dendrobium species are closely related and the Guizhou Dendrobium has rich genetic diversity.Con-clusion ISSR and SSR molecular markers can be effectively used in the genetic diversity analysis of Dendrobium.The genetic var-iation of Dendrobium is rich and can be applied in the crossbreeding of excellent germplasm resources of Dendrobium adapted to the environment of Guizhou.
