电针对脑卒中肢体痉挛大鼠皮质神经细胞铁死亡的影响

目的 观察电针对脑卒中肢体痉挛(PSS)大鼠皮质神经细胞超微结构、亚铁离子(Fe2+)、丙二醛(MDA)、谷胱甘肽(GSH)表达的影响,探讨电针治疗脑卒中肢体痉挛中铁死亡的机制。方法 采用改良ZeaLonga线栓+内囊注射NMDA法制作脑PSS大鼠模型。将30只健康成年雄性SD大鼠随机分为假手术组,模型组及电针组。电针组取穴患侧"阳陵泉、曲池",1次/日,30min/次,连续7d。记录Zea-longa神经功能评分、改良Ashworth肌张力评分,透射电镜观察线粒体变化,普鲁士蓝染色观察皮质铁离子聚集情况,生化试剂盒检测大鼠皮质Fe2+、MDA含量,ELISA检测大鼠皮质GSH含量。结果 Zea-longa神经功能评分:与模型组相比,电针组评分降低(P<0。05);改良Ashworth肌张力评分:与模型组相比,电针组评分降低(P<0。05);透射电镜:与模型组相比,电针组皮质神经细胞损伤减轻,线粒体萎缩程度降低,线粒体嵴更为明显。普鲁士蓝染色:与模型组相比,电针组皮质铁聚集明显减少。实验室检测结果:与模型组相比,电针组Fe2+、MDA含量降低(P<0。01),GSH含量升高(P<0。01)。结论 电针可改善脑卒中肢体痉挛,其作用机制可能与电针抑制皮质神经细胞铁死亡,发挥脑保护作用,促进神经功能恢复有关。
Effect of Electroacupuncture on Ferroptosis in Cortical Nerve Cells of Rats with Post Stroke Spasticity
Objective To observe the effects of electroacupuncture on the ultrastructure,ferrous ion(Fe2+),malondialdehyde(MDA),and glutathione(GSH)expression in cortical neuronal cells of rats with post stroke spasticity(PSS),exploring the mechanisms of ferroptosis in electroacupuncture for post stroke spasticity.Methods A rat model of cerebral PSS was produced u-sing the modified ZeaLonga thread bolus+internal capsule injection of NMDA method.Thirty healthy adult male SD rats were randomly divided into sham surgery group(SS group),model group(M group)and electroacupuncture group(EA group).The electroacupuncture group took points on the affected side"Yanglingquan and Quchi",1 time/day,30min/time,for 7d.Zea-lon-ga Neurological Function Score and Modified Ashworth Dystonia Score were recorded,mitochondrial changes were observed by transmission electron microscopy,cortical iron ion aggregation was observed by Prussian blue staining,and cortical Fe2+and MDA contents were detected by biochemical kits,and cortical GSH contents were detected by ELISA in rats.Results Zea-longa Neurological Function Score:compared with the model group,the score of the electroacupuncture group was reduced(P<0.05);Modified Ashworth Dystonia Score:compared with the model group,the score of the electroacupuncture group was reduced(P<0.05);Transmission electron microscopy:compared with the model group,the damage to the cortical neuronal cells of the elec-troacupuncture group was reduced,the degree of mitochondrial atrophy was reduced,and mitochondrial cristae were more obvi-ous.Prussian blue staining:compared with the model group,cortical iron aggregation was significantly reduced in the electroacu-puncture group.Laboratory test results:compared with the model group,Fe2+and MDA content decreased in the electroacupunc-ture group(P<0.01),and GSH content increased(P<0.01).Conclusion Electroacupuncture can improve post stroke spas-ticity,and its mechanism of action may be related to electroacupuncture inhibiting ferroptosis of cortical nerve cells,exerting cere-bral protective effects and promoting neurological recovery.
