基于UPLC-Q-TOF-MS技术探讨桔梗对芩百清肺浓缩丸在肺组织中化学成分的影响

目的 比较芩百清肺浓缩丸全方和缺桔梗方在大鼠肺组织中化学成分的差异,探讨桔梗在该复方制剂中的引经作用.方法 灌胃给予大鼠芩百清肺浓缩丸全方和缺桔梗方,采集肺组织处理后,运用UPLC-Q-TOF-MS技术,在正、负离子扫描模式下进行测定.运用MarkerView 1.2.1软件进行主成分分析,找出组间样品中的差异离子,结合PeakView 2.0/Masterview 1.0软件,依据保留时间、一级二级质谱图、元素组成等信息进行目标筛查,确定两组间的差异化合物,进行相对定量分析.结果 在两组方成分入肺的比较中,全方组共找到8个相对浓度较高的化合物,具有显著性差异.结论 桔梗作为引经药能够增加芩百清肺浓缩丸中有效成分在肺组织的分布.
Effects of Platycodonis Radix on the chemical constituents of Qinbai Qingfei Concentrated Pills in the lung tissues by UPLC-Q-TOF-MS
Objective To make a comparison of changes of constituents in the lung tissues between Qinbai Qingfei Concentrated Pills(QQCP)and lacked Platycodonis Radix of QQCP to explore the effect of Platycodonis Radix as a channel ushering drug in the Chinese herbal compound QQCP.Methods Rats were given intragastric administration of QQCP and lacked Platycodonis Radix of QQCP,and then collected the lung tissues.Using the technology of UPLC-Q-TOF-MS to carry out the determination in posi-tive and negative ion scanning mode.Find the different ions in the samples between the groups by principal component analysis u-sing the software of MarkerView 1.2.1.Meanwhile,identify different compounds and analyse auantivatively on the basis of infor-mation,including retention time,first order mass spectrum,second order mass spectrum and elementary composition combined with the software of PeakView 2.0/masterview 1.0.Results In the comparion of chemical component absorbed into the lung tissues,eight relatively higher compounds with significant differences were found between two groups.Conclusion Platycodonis Radix could increase the distribution of effective constituent of QQCP in lung tissues as a channel ushering drug.
