Effect of Radix Angelica Sinensis and Radix Hedysari Ultrafiltration on Myocardial Fi-brosis and HIF-1α、OPN Protein in Rats with Coronary Heart Disease
Objective To investigate the effect of Radix Angelica Sinensis and Radix Hedysari Ultrafiltration on myocardial fibro-sis and HIF-1 α and OPN protein in rats with coronary heart disease induced by high fat diet.Methods will Thirty-six Wistar male rats were randomly divided into blank group,model group,Rosuvastatin calcium tablet group(0.1mg·kg-1·d1),Radix Angelica Sinensis and Radix Hedysari Ultrafiltration low-dose group(0.1g·kg-1·d-1),Radix Angelica Sinensis and Radix Hedysari Ultrafiltration low-medium dose group(0.3g·kg-1·d-1),and Radix Angelica Sinensis and Radix Hedysari Ultrafil-tration high-dose group(0.5g·kg-1·d-1).After 4 weeks of drug intervention,the expression levels of serum CK-MB and cTnI were detected by ELISA,the changes of myocardial fibrosis were observed by Masson staining,the expressions of TNF-α,MMP-9 and TGF-β1 proteins in myocardial tissue were detected by immunohistochemistry,and the expressions of HIF-1αand OPN proteins in myocardial tissue were detected by immunofluorescence.Results Compared with the blank group,the fibro-sis area and degree of the model group were significant,and the fibrosis area and degree of the Radix Angelica Sinensis and Radix Hedysari Ultrafiltration group were reduced.Compared with the blank group,the protein expressions of TNF-α,MMP-9 and TGF-β1 in the model group were increased,while the protein expressions of TNF-α,MMP-9 and TGF-β1 in the Radix An-gelica Sinensis and Radix Hedysari Ultrafiltration group were decreased compared with the model group.Compared with blank group,HIF-1α expression in model group was significantly decreased(P<0.01)and OPN expression was significantly in-creased(P<0.01).Compared with model group,HIF-1α expression was increased(P<0.05)and OPN expression was de-creased(P<0.05)in medium and high dose groups of Radix Angelica Sinensis and Radix Hedysari Ultrafiltration.Conclusion Radix Angelica Sinensis and Radix Hedysari Ultrafiltration can improve the expression of HIF-1 α and OPN protein in coronary heart disease rats induced by high fat diet.
