Study on the mechanism of VD neuroinflammation by regulating microglial polarization
Objective The effect of Yifei Xuanfei Jiangzhuo formula on microglial polarization and regulation of TLR4/NF-KBsignaling pathway in hippocampal tissues of rats with vascular dementia was observed.Methods 75 mice were randomlydivid-ed into sham surgery group(SHAM group),model group(MOD group),low-dose group(LDY group),medium-dose group(MDY group),high-dose group(HDY group),and piracetam group(PIR group).The 2-VO method(bilateral common ca-rotid artery permanent ligation)was used to test the learning and memory behavior of rats by water maze before and after model-ing.HE staining and fluoroelectron microscopy analysis of hippocampal pathological morphology;TUNEL method to detect apopto-sis in hippocampal CA1 region,immunofluorescence double-standard staining to detect microglia polarized phenotypic proteins CD16/32+/Iba-1+and CD206+/Iba-1+co-standardization,RT-PCR to detect gene expression of microglia M1/M2 secre-tion of proactive/anti-inflammatory factors TNF-α,IL-1β,IL-4 and TGF-B,western blot to analyze microglia polarized iNOS and Arginase-1 and TLR4/NFKB signaling pathway-related protein expression.Results Compared with the model group,the ratio of prohibitive lung xuan lung turbidity reduction formula to apoptosis and neuronal damage in hippocampal tissue was reduced(P<0.05),among which the best effect was at high doses(P<0.01),and the number of microglia M1 CD16/32 positive cells decreased(P<0.05),which could be reversed by yifei xuanfei jianzhuo decoction.Conclusion The neuroprotec-tive effect of Yilungxuan lung turbidity decoction protects neurons by inhibiting inflammation-related TLR4/NFKB signaling path-way,promoting the transformation of microglia to M2 type,and improving vascular dementia.
