Based on RoRty/Foxo3 immmune imbalance to explore the effect of Yangyin Qingfei De-coction Plus on the immune microenvironment in Mycoplasma infected mice
Objective Based on the RORγt/Foxp3 immune imbalance mechanism,we investigated the dynamic correlation be-tween the dose-effect relationship of traditional Chinese medicines and the immune microenvironment of MP-infected mice by observing the influences of different doses of Yangyin Qingfei Decoction Plus to the immune microenvironment.Methods Thirty-six BALB/c mice were randomly divided into normal group,model group,aci group and YQDP group(high,middle and low dose groups).Except for the normal group,all groups of mice were anesthetized and nasal drops of MP bacterial solution at 1010 CCU/ml were used to construct the model,and drug intervention was started after successful modeling and sampling was per-formed on the 7th day after the drug intervention.The ultrastructural changes in lung tissue epithelium of mice were observed,the mRNA content of RORγt and Foxp3 in lung tissues of mice was measured by qPCR,and the protein expressions of RORγt and Foxp3 in lung tissue were analyzed by Western Blot and immunohistochemistry.The determination of DPPC in mice BALF was measured by HPLC,and the levels of TGF-β,IL-17A,IL-10 in serum and SP-A,SP-D in BALF of mice were measured by ELISA.Results Infection increased expression of RORγt and decreased expression of Foxp3 in lung tissue,and increased ser-um levels of TGF-βand IL-17 A,and decreased serum and BALF levels of IL-10,SP-A,SP-D and DPPC(P<0.05).After treatment,RORγt,TGF-β and IL-17 A were decreased in mice,and the expressions of Foxp3,IL-10,SP-A,SP-D and DPPC were rebounded to varying degrees.Conclusion Yangyin Qingfei Decoction Plus can affect the dynamic changes of im-mune microenvironment in mice lung by regulating the RORγt/Foxp3 immune homeostasis mechanism and the dynamic differential expressions of lung surface active substances SP-A,SP-D and DPPC.
