Exploring the effects of traditional Chinese medicine Guasha on the skin tissues of rats with lumbar intervertebral disc herniation based on HSP70
Objective The analgesic effect of Guasha on rats was observed in lumbar disc herniation(LDH)model rats to explore whether it affects the skin chakra and promotes local skin blood circulation by promoting the expression of HSP70,thus relieving the chakra occlusion and regulating dorsal root ganglion injury to achieve analgesic effect.Methods Forty SD male rats were ran-domly divided into sham group,model group,Guasha group and sham-Guasha group,with 10 rats in each group.Autologous myeloid transplantation was used to establish the LDH disease model,and Guasha intervention was given to the Dumai and bilater-al bladder meridian to observe its effects on the mechanical pain threshold(PWT)of their right hindfoot,local skin temperature,serum TGF-β1,dorsal root ganglion(DRG)and local skin morphology of the back,and the local skin tissues of HSP70 mRNA and proteins.Results Compared with the model group,PWT was significantly higher in the Guasha group(P<0.05),but there was no statistically significant difference in the sham-Guasha group(P>0.05);compared with the non-Guasha groups,such as the sham group,the model group,and the sham-Guasha group,local skin temperature of the back of the rats after Guasha was significantly higher(P<0.05);compared with the sham group,serum TGF-β1 level of the model group was significantly lower(P<0.05),the TGF-β1 level in the gua sha group was significantly higher than that in the model and sham-Guasha groups(P<0.05);the DRG morphology of the rats in the model and sham-Guasha groups showed obvious signs of inflammatory edema and injury compared to the sham group,whereas the nerve injury and inflammatory infiltration in the Guasha group were re-duced;capillary dilatation was visible in the dermal layer of the skin of the Guasha group compared to the non-gua sha groups.The dermis of the skin in the Guasha group showed dilated capillaries,scattered distribution of abundant erythrocytes,and partial infiltration of immune cells such as neutrophils and lymphocytes;compared with the non-Guasha group,the expression of HSP70 mRNA and protein in the Guasha group was significantly increased(P<0.05).Conclusion Guasha can reduce the inflammato-ry response of the body and alleviate the pain response triggered by LDH,and its mechanism may be related to elevating the local skin temperature,stimulating the skin complexes,mobilizing the cascade response of the body,promoting the local blood circula-tion of the skin,and alleviating the stagnation caused by the blockage of the veins and collaterals,among which the HSP70 protein may be a key target protein of the activation of the cascade response of the body in Guasha.
