目的 探讨当归芍药散对子宫内膜异位症(EMs)模型大鼠异位内膜上皮间充质转化相关因子的影响。方法 采用大鼠自体子宫内膜移植法构建EMs模型大鼠。将造模成功的48只EMs模型大鼠随机分为假手术组、模型组、当归芍药散低、中、高剂量组(3。96,7。92,15。84g·kg-1)、西药组(孕三烯酮胶囊0。25 mg·kg-1),每组8只。假手术组和模型组大鼠给予生理盐水(10mL·kg-1)灌胃。采用苏木素-伊红(HE)染色法对正常内膜及异位内膜进行组织形态学观察;免疫组化法检测正常子宫内膜与异位内膜组织的E-cadherin、N-cadherin蛋白表达;蛋白免疫印迹法(Western blot)检测正常内膜及异位组织β-catenin蛋白表达。结果 与假手术组正常内膜相比,EMs各组异位子宫内膜组织的E-cadherin蛋白表达下降而β-catenin、N-cadherin蛋白含量升高(P<0。05)。与模型组相比,当归芍药散各剂量组的E-cadherin蛋白上调而β-catenin、N-cadherin蛋白表达含量下降(P<0。05)。结论 当归芍药散对EMs模型大鼠的治疗作用可能与调节上皮间充质转化相关因子β-catenin以及E-cadherin、N-cadherin的表达有关。
Effect of Danggui Shaoyao Decoction on the factors related to epithelial mesenchymal transformation in Endometriosis model rats
Objective To explore the effect of Danggui Shaoyao Decoction on the factors related to epithelial mesenchymal trans-formation of Endometriosis(EMs)model rats.Methods Construct an EMs rat model using autologous endometrial transplantation in rats.48 rats were randomly divided into a Sham surgery group,a model group,a low,medium and high dose group of Danggui Shaoyao Decoction(3.96,7.92,15.84g·kg-1),and a western medicine group(0.25mg·kg-1 for Gestrinone capsules),with 8 rats in each group.Rats in the Sham surgery group and the model group were given normal saline(10mL·kg-1)by ga-vage.Using hematoxylin eosin(HE)staining method to observe the histological morphology of normal and ectopic endometrium;Immunohistochemical staining was used to detect the expression of E-cadherin and N-cadherin proteins in normal and ectopic endometrium tissues;Western blot detection of normal endometrium and ectopic tissues β-catenin protein expression.Re-sults Compared with the normal endometrium in the Sham surgery group,the expression of E-cadherin protein in ectopic endo-metrium in the EMs group decreased the protein content of β-catenin and N-cadherin increased(P<0.05).Compared with the model group,the E-cadherin protein in each dose group of Danggui Shaoyao Decoction was upregulated the expression levels of β-catenin and N-cadherin proteins decreased(P<0.05).Conclusion The therapeutic effect of Danggui Shaoyao Decoction on EMs model rats may be related to regulating the expression of epithelial mesenchymal transition related factors β-catenin,E-cadherin and N-cadherin.
