Genetic diversity analysis of Trametes versicolor germplasm resources based on RAPD and ISSR markers and establishment of SCAR markers
To investigate the genetic background of wild Trametes versicolor germplasm resources,18 strains from different ori-gins were identified and analyzed for genetic diversity based on two molecular marker technologies,RAPD and ISSR,and a more stable SCAR marker was also established.The results showed numerous bands had been amplified by the primers of RAPD and IS-SR,with polymorphism rates of 100%and 97.4%,respectively.This indicated RAPD and ISSR successfully detected the abun-dant genetic diversity of the tested strains and effectively distinguished the T.versicolor strains.The NPL,PPB,Na,Ne,He,and I values of the T.versicolor populations from Changsha(CS)and Ningxiang(NX)in the south were similar and higher,while those from Shandong,Liaoning and Jilin in the north were lower.The clustering results illustrated there might be a cluste-ring pattern from south to north.The SCAR markers obtained also might be associated with the gene related to the edge thickness of the fruiting bodies of T.versicolor.This study has improved the molecular markers of the genetic material of T.versicolor,and provided a certain research basis for subsequent breeding of new varieties and regulation of trait-related genes.
