Protective effect of Panax notoginseng rhizomes on acute kidney injury induced by ische-mia-reperfusion
Objective To investigate the protective effect of Panax notoginseng rhizomes on renal ischemia-reperfusion injury(RIRI).Methods Male SD rats were randomly divided into a sham group,a model group,and a Panax notoginseng rhizomes group,and a bilateral RIRI model was established after pretreatment administration for 7 days,and blood and both kidneys of rats were collected after 24 hours of reperfusion.Blood samples were utilized for the determination of serum creatinine(SCr),blood urea nitrogen(BUN),monocyte(MONO)and neutrophil(NEUT)levels to assess renal function.Kidney tissue was used for the determination of malondialdehyde(MDA)content to detect oxidative stress;HE staining was carried out to observe the pathologic changes in renal tissues;and immunofluorescence staining with CD86 was performed to evaluate the pro-inflammatory macro-phage infiltration of renal tissues.Results Compared with the sham group,the model group had increased levels of SCr,BUN,MONO,NEUT,and MDA(P<0.001),increased pathologic changes in renal tubular morphology and injury scores(P<0.001),and increased severity of pro-inflammatory macrophage infiltration and number of CD86-positive cells(P<0.001).Compared with the model group,the Panax notoginseng rhizomes group significantly reduced SCr,BUN,MONO,NEUT,and MDA levels(P<0.01),reduced renal tubular morphopathologic damage and scores(P<0.001),and reduced pro-inflammatory macrophage infiltration and CD86-positive cell count(P<0.001).Conclusion Panax notoginseng rhizomes can protect kidney tissue by anti-inflammation and anti-oxidation,reduce ischemia-reperfusion injury,and inhibit the infiltration of CD86 pro-inflammatory macrophages.
