基于粪便代谢组学研究健脾化浊方对动脉粥样硬化小鼠的影响

目的 基于液相色谱-质谱联用(LC-MS)研究健脾化浊方对动脉粥样硬化(AS)小鼠粪便代谢物的调节机制。方法 采用高胆碱饮食建立AS动物模型,观察小鼠主动脉斑块病理变化,运用LC-MS技术分析小鼠粪便样品代谢轮廓,结合主成分分析(PCA)、正交最小偏二乘判别分析(OPLS-DA)等手段鉴定并筛选出小鼠粪便差异代谢物及代谢通路。结果 健脾化浊方可抑制小鼠AS斑块形成,调节血清脂质水平,并回调77种与AS相关的潜在生物标志物、10种关键生物标志物以及8条代谢通路(维生素B6代谢、硫胺素代谢、类固醇生物合成、甘油磷脂代谢、嘌呤代谢、精氨酸和脯氨酸代谢、初级胆汁酸生物合成、类固醇激素生物合成)。结论 健脾化浊方发挥治疗AS的作用机制可能是通过调节AS小鼠维生素代谢、核苷酸代谢、脂质代谢以及氨基酸代谢等过程来实现。
Study on the effect of Jianpi Huazhuo Recipe on atherosclerosis mice based on fecal meta-bonomics
Objective Based on liquid chromatography-mass spectrometry(LC-MS),the regulatory mechanism of Jianpi Huazhuo Recipe on fecal metabolites in atherosclerotic(AS)mice was studied.Methods The AS animal model was established by high choline diet,and the pathological changes of aortic plaque in mice were observed.The metabolic profile of mouse feces samples was analyzed by LC-MS technology,and the differential metabolites and metabolic pathways in mouse feces were identi-fied and screened by means of principal component analysis(PCA)and orthogonal least squares discriminant analysis(OPLS-DA).Results Jianpi Huazhuo Recipe can inhibit the formation of AS plaque in mice,regulate the serum lipid level,and recall 77 potential biomarkers,10 key biomarkers and 8 metabolic pathways related to AS(vitamin B6 metabolism,thiamine metabo-lism,steroid biosynthesis,glycerophosphate metabolism,purine metabolism,arginine and proline metabolism,primary bile acid biosynthesis and steroid hormone biosynthesis).Conclusion The mechanism of Jianpi Huazhuo Recipe in treating AS may be through the regulation of vitamin metabolism,nucleotide metabolism,lipid metabolism and amino acid metabolism in AS mice.
