Effects of electroacupuncture on mGLuR5 in MPTP-induced PD model mice
Objective To observe the effects of electroacupuncture at Fengfu and Taichong points on metabolic glutamate 5(mGluR5)and its pathway in mice with Parkinson's disease(PD),and to explore the possible mechanism of electroacupuncture in the treatment of PD.Methods C57BL/6 mice were randomly divided into normal group(Ctrl),model group(MPTP),inhibi-tor group(CHPG),inhibitor plus electroacupuncture group(CHPG+EA),and electroacupuncture group(EA group),with 10 mice in each group.The PD model was replicated by continuous intraperitoneal injection of MPTP for 7 days.Electroacupuncture was applied to Fengfu and Taichong points for 30 min/d once/d for 12 consecutive days.Behavioral changes of mice in each group were observed.mGluR5 expression was detected by immunohistochemistry;mGluR5,p-ERK,p-CREB and BDNF protein ex-pression in substantia nigra was detected by Western blot,and mRNA expression levels of mGluR5,ERK,CREB and BDNF were detected by PCR.Results Compared with the Ctrl group,mice in the MPTP,CHPG,and CHPG+EA groups showed behavioral changes(P<0.01),while mice in the EA group showed improved behavior(P<0.01).nigral mGluR5-positive expression increased in mice in the MPTP,CHPG,and CHPG+EA groups(P<0.01),while nigral mGluR5 positive expression decreased(P<0.01).mRNA relative expression and protein expression of mGluR5 in MPTP,CHPG,and CHPG+EA groups increased(P<0.01),and mRNA relative expression and protein expression of mGluR5 in EA group was down-regulated(P<0.01).ERK1/2 in MPTP,CHPG,and CHPG+EA groups,CREB and BDNF were down-regulated(P<0.01),and the relative mR-NA expression and protein expression of ERK1/2,CREB and BDNF were up-regulated(P<0.01)in the EA group.Conclu-sion Electroacupuncture at Fengfu and Taichong points improved the motor dysfunction of PD mice and inhibited the activation of mGluR5 pathway in the substantia nigra of the midbrain.
