Effects of Loganetin on proliferation of white blood cells and CDC20/MAD2 expression
Objective To investigate the effects of loganetin on Cdc20 and Mad2 during the proliferation of human acute myeloid leukemia MOLM-13.Methods The proliferation of MOLM-13 cells was detected by CCK-8 method after 24,48 and 72 h treatment with different concentrations of loganetin.Flow cytometry was used to detect the changes of cell cycle and apoptosis of MOLM-13 cells after 24h treatment with different concentrations of loganetin.Western blot analysis of Mad2 and Cdc20 protein expression in MOLM-13 cells treated with different concentrations of loganetin for 48h.Results Loganetin could significantly in-hibit the proliferation of MOLM-13 cells,Promote the apoptosis of MOLM-13 cells,arrest the cell cycle in G2/M phase,and decrease the expression levels of Mad2 and Cdc20 Protein.Conclusion Loganetin has a significant inhibitory effect on the prolif-eration of MOLM-13 cells in human acute myeloid leukemia,and the mechanism may be related to regulating the expression lev-els of Mad2 and Cdc20 protein and influencing cell mitosis.
