Effect of Rhubarb Lingxian on expression of IL-6 and EGR-1 related proteins in rats with bile duct inflammation
Objective To explore the mechanism of Dahuang Lingxian Recipe on the formation of intrahepatic bile duct stones on the ba-sis of constructing the model of hepatobiliary inflammation in SD rats.Methods Forty male SD rats were randomly divided into blank group,lipopolysaccharide model group,ursodeoxycholic acid capsule control group and rhubarb Lingxian formula experimental group,with 10 rats in each group.The normal group was fed with conventional feed;The model group,the control group and the experimental group were injected with 1.25mg/kg lipopolysaccharide in the common bile duct at one time.At the same time,the control group was given ursodeoxycholic acid and the experimental group was given Dahuang Lingxian formula for 7 days.Blood was taken for ELISA,liver tissue was stained with he,and WB was used to detect the expression of IL-6 and Egr-1 protein.Results Compared with the other three groups,the pathological morphology of liver in the model group showed that the degree of inflammatory infiltration was obvious;There was significant difference in the expression of IL-6 and Egr-1 protein in blood and liver of rats in each group(P<0.01);Compared with the normal group,the expression of IL-6 and Egr-1 protein in blood and liver of the model group and the control group were significantly higher(P<0.01);Compared with the normal group,there was no significant difference in the expression of IL-6 and Egr-1 protein in blood and liver of the experimental group(P>0.05).Conclusion Dahuang Lingxian granule may inhibit the re-lease of inflammatory factors such as IL-6 and Egr-1 in cholangitis rat model,reduce liver injury and make it tend to normal level,so as to block the formation of stones.
