Effects of Zhizhu Wan modulating VIP,cAMP,PKA,AQP1 on Intestinal Fluid Metabolism of Slow Transit Constipation Mice
Objective To investigate the effects of Zhizhu Wan(ZZW)on colon Motility and AQP1 in slow transit constipation(STC)mice.Methods Mice of slow transit constipation with spleen deficiency was established by senna gavage+hunger and satiety disorders method and randomly divided into the model group(distilled water),mosapride group(2.5 mg·kg-1),ZZW group(11.7 g·kg-1),and a normal group(distilled water),with 10 mice in each group.After 7 days of treatment,observe the general behavioral characteris-tics of mice in each group,detect the intestinal propulsive rate and fecal water content of mice in each group,observe the pathological changes of colon tissue of mice in each group by HE staining,detect the content of VIP,cAMP,PKA in the colon of mice by Elisa meth-od,and detect the expressions of cAMP,PKA,and AQP1 protein in colon of mice by Western-blotting method.Results Compared with the normal group,the intestinal propulsive rate and fecal water content were reduced in the model group(P<0.05),Elisa method show that VIP,cAMP and PKA contents in colon tissues were reduced(P<0.05 or P<0.01),and the results of Western-Blot show that cAMP and PKA protein expression in colon tissues was reduced(P>0.05,P<0.01),and AQP1 protein expression was increased(P<0.05).Compared with the model group,the intestinal propulsive rate of mosapride group and ZZW group show an increasing trend(P>0.05),and the water content of feces was increased(P<0.05).Elisa method show that the VIP of colon tissues showed an increasing trend(P>0.05),and the content of cAMP and PKA were significantly increased(P<0.01).Western-Blot results show that the ex-pression of cAMP and PKA proteins in colon tissues increased(P>0.05,P<0.01),and the expression of AQP1 proteins decreased(P<0.05).Conclusion Therefore,it is hypothesized that ZZW pills can improve constipation symptoms in STC mice with spleen defi-ciency,and the mechanism may be related to the increase of VIP,cAMP,and PKA content and thus down-regulation of colonic AQP1 expression,decrease of water reabsorption to soften stools.
