Danshou Decoction reduces endothelial cell damage by regulating miR-744-3p to relieve re-current spontaneous abortion
Objective To observe the effect of Danshou Decoction on regulating miR-744-3p,reducing endothelial cell damage and alleviating recurrent spontaneous abortion and its potential molecular mechanism.Methods The placental tissues of mice in the recurrent spontaneous abortion model group and the Danshou Decoction group were collected,and then we weighed the embryo and calculated the embryo absorption rate.The expression of miR-744-3p in the tissues was detected by Real-time PCR.MiR-744-3p mimics or in-hibitors were transfected into human umbilical vein endothelial cells.The effects of miR-744-3p on the proliferation,migration and tu-bule formation of endothelial cells were detected by CCK-8,scratch and tubule formation test.Real-time PCR and Western blot were used to detect the effect of miR-744-3p on TLR4 expression,and double luciferase reporter gene assay was used to verify the targeting of miR-744-3p to TLR4.Results Compared with the model group,Danshou Decoction could significantly inhibit the embryo absorp-tion rate and up-regulate the expression of miR-744-3p in recurrent abortion mice.Overexpression of miR-744-3p significantly enhanced the proliferation,migration and tubular formation of endothelial cells compared with the control group,while inhibition of miR-744-3p showed the opposite results.MiR-744-3p can inhibit TLR4 expression by directly targeting TLR4.Conclusion Danshou decoction can inhibit the expression of TLR4 by inducing the expression of miR-744-3p,thus reducing endothelial cell damage and al-leviating the process of recurrent spontaneous abortion.
