平喘颗粒对卵清蛋白致敏诱导的哮喘小鼠巨噬细胞M2极化及补体因子D的影响

目的 观察平喘颗粒对哮喘小鼠巨噬细胞M2极化和补体因子D(CFD)表达的影响,探讨其防治哮喘气道重塑可能的作用机制。方法 将40只雌性C57BL/6小鼠随机分为空白组、模型组、布地奈德组及平喘颗粒组,每组10只。除空白组外,其余3组通过卵清蛋白(OVA)腹腔注射及雾化构建模型。造模成功后,分别给予相应药物治疗,治疗4周后取材。Masson染色观察小鼠肺组织病理改变情况,ELISA检测支气管肺泡灌洗液(BALF)和血清中IL-4的水平,流式细胞术检测M1型巨噬细胞(F4/80+CD86+)及M2型巨噬细胞(F4/80+CD206+)表达水平,Western blot检测肺组织中FIZZ1、Ym1、CFD和α-SMA的表达,免疫荧光双染色检测肺组织F4/80和CFD的表达。结果 与空白组相比,模型组显示大量胶原纤维沉积,BALF和血清中IL-4的水平明显升高,肺组织M1和M2型巨噬细胞极化百分比明显升高,肺组织中FIZZ1、Ym1、CFD和α-SMA蛋白表达量明显升高,F4/80与CFD存在共表达,F4/80和CFD的荧光强度明显增强(P<0。01);与模型组相比,平喘颗粒可减少胶原纤维沉积,明显降低BALF和血清中IL-4的水平、肺组织M2型巨噬细胞极化百分比,肺组织中FIZZ1、Ym1、CFD和α-SMA蛋白表达量也明显降低,F4/80和CFD的荧光强度减弱(P<0。01)。结论 平喘颗粒可能通过降低CFD表达,从而抑制巨噬细胞M2极化,达到治疗哮喘气道重塑的目的。
Effects of Pingchuan Granules on M2 polarization,complement factor D expression and air-way remodeling of macrophages in asthmatic mice
Objective To observe the effects of Pingchuan granule on the M2 polarization of macrophage and the expression of comple-ment factor D in asthmatic mice,and to explore its possible mechanism of prevention and treatment of airway remodeling in asthma.Methods Forty female C57BL/6 mice were randomly divided into blank group,model group,budesonide group and Pingchuan granule group,with 10 mice in each group.Except for the blank group,the other three groups were modeled by intraperitoneal injection and Aerosol inhalation of OVA.After successful modeling,they were given corresponding medications,and the materials were taken after 4 weeks of treatment.The pathological changes of mice lung tissue were observed by Masson staining;ELISA was used to detect the levels of IL-4 in BALF and serum,Flow Cytometry was used to detect the expression levels of M1 macrophages(F4/80+CD86+)and M2 macrophages(F4/80+CD206+),Western blot method was used to detect the expression of FIZZ1,Ym1,CFD and α-SMA in lung tis-sue,and Immunofluorescence double staining was used to detect the expression of F4/80 and CFD in lung tissue.Results The experi-mental study showed that compared with the blank group,the model group showed a large amount of collagen fiber deposition,the level of IL-4 in BALF and serum was significantly increased,the percentage of polarization of M2 and M1 macrophages in lung tissue was signif-icantly increased,the expression of FIZZ1,Ym1,CFD and α-SMA protein in the lung tissues was significantly increased,and there was co-expression of F4/80 and CFD,and the fluorescence intensity of F4/80 and CFD was significantly enhanced(P<0.01);com-pared with the model group,Pingchuan granule could reduce collagen fiber deposition,significantly reduce the level of IL-4 in BALF and serum,the percentage of the M2 polarization of macrophage in lung tissue,the expression of FIZZ1,Ym1,CFD and α-SMA pro-tein in lung tissues was also significantly reduced,and decreased fluorescence intensity of F4/80 and CFD(P<0.01).Conclusion-Pingchuan granule may reduce the expression of CFD,thereby inhibiting the M2 polarization of macrophage,and finally achieve the pur-pose of treating asthma airway remodeling.
