摘要
目的 研究附子-干姜含药血清对缺氧/复氧损伤的H9C2细胞保护作用.方法 通过给药大鼠制备附子干姜含药血清,体外H9C2细胞建立缺氧/复氧损伤模型.细胞分组为Control组(Con)、Hypoxia-reoxygenation组(H/R)、FG含药血清组(FG);CCK-8法检测各组细胞活力;流式细胞术检测各组细胞凋亡率及ROS;试剂盒检测各组H9C2细胞中SOD、MDA、TNF-α、IL-10的表达;RT-PCR和WB分别检测各组细胞中TRPV1、CGRP基因和蛋白的表达.结果 FG可提高H/R损伤的H9C2细胞存活率(P<0.01),降低MDA、TNF-α、IL-10的表达及凋亡率、ROS,提高SOD的表达并促进TR-PV1和CGRP基因和蛋白的表达.结论 FG能够通过激活TRPV1/CGRP信号通路提高H/R损伤的H9C2细胞存活率,降低氧化损伤和炎症损伤发挥治疗作用.
Abstract
Objective Study the protective effect of Fuzi Ganjiang serum(FG)on H9C2 cells after hypoxia/reoxygenation(H/R)inju-ry.Methods FG medicated serum was prepared from rats,and H9C2 cells were used to establish H/R injury model in vitro.H9C2 cells were divided into Control group(Con),H/R group and FG group.The cell viability was detected by CCK-8 method,the apoptosis rate and ROS were detected by flow cytometry,the expressions of SOD,MDA,TNF-α and IL-10 were detected by kit,the gene and pro-tein expressions of TRPV1 and CGRP were detected by RT-PCR and WB,respectively.Results FG could improve the survival rate of H9C2 cells after H/R injury(P<0.01),decreased the expression of MDA,TNF-α,IL-10 apoptosis,ROS.increased the expres-sion of SOD,and increased the gene and protein expression of TRPV1 and CGRP.Conclusion FG can improve the survival rate of H9C2 cells after H/R injury by activating TRPV1/CGRP signaling pathway,and reduce oxidative damage and inflammatory damage.
