基于细胞焦亡途径探讨清金通络颗粒对肺炎支原体肺炎的干预机制

目的 探讨清金通络颗粒通过NLRP3/Caspase-1/GSDMD细胞焦亡途径对肺炎支原体肺炎(MPP)小鼠干预机制。方法 将60只Balb/c小鼠随机分为空白组、模型组、清金通络颗粒组,MCC950(NLRP3抑制剂)组,清金通络颗粒+MCC950组,阿奇霉素组。采用MP菌株滴鼻法建立MPP小鼠模型。造模后予药物治疗,治疗14d后取材。HE染色观察小鼠肺组织病理改变,ELISA法检测IL-1β、IL-18含量,Western blot法检测NLRP3、caspase-1、ASC、GSDMD蛋白表达量,PCR法检测小鼠肺组织中NLRP3、caspase-1、ASC、GSDMD、IL-1β、IL-18 mRNA水平。结果 与空白组比较,模型组小鼠肺组织出现病理损伤,血清与肺组织中IL-1β、IL-18含量升高,肺组织中NLRP3、caspase-1、ASC、GSDMD蛋白表达水平升高,肺组织中NLRP3、caspase-1、ASC、GSDMD、IL-1β、IL-18 mRNA表达升高(P<0。05)。与模型组相比,其余4组肺组织病理损伤得到不同程度改善,血清与肺组织中IL-1β、IL-18含量均下降,肺组织中NLRP3、caspase-1、ASC、GSDMD蛋白表达水平均下降,肺组织中NLRP3、caspase-1、ASC、GSDMD、IL-1β、IL-18 mRNA表达均下降(P<0。05)。结论 清金通络颗粒可能通过调控NLRP3/Caspase-1/GSDMD介导的细胞焦亡,改善MPP小鼠肺部炎症。
Exploring the Mechanism of Qingjin Tongluo Granules in intervening Mycoplasma Pneumoni-ae Pneumonia Based on the Pathway of Cell Pyroptosis
Objective To explore the intervention mechanism of Qingjin Tongluo Granules on Mycoplasma Pneumonia Pneumonia(MPP)mice through NLRP3/Caspase-1/GSDMD cell pyroptosis pathway.Methods Sixty Balb/c mice were randomly divided into blank group,model group,Qingjin Tongluo Granule group,MCC950(NLRP3 inhibitor)group,Qingjin Tongluo Granule+MCC950 group,and Azithromycin group.A mouse model of MPP was established using the nasal drip method of Mycoplasma Pneumoniae(MP)strain.The rats were treated with drugs after modeling,and the materials were taken after 14 days of treatment.The pathological changes of mouse lung tissues were observed by HE staining.The contents of IL-1β and IL-18 were detected by ELISA.The protein expres-sions of NLRP3,caspase-1,ASC and GSDMD were detected by Western blot.The mRNA levels of NLRP3,caspase-1,ASC,GSD-MD,IL-1β and IL-18 in mouse lung tissues were detected by PCR.Results Compared with the blank group,the model group had pathological injury in lung tissue,increased IL-1 β and IL-18 contents in serum and lung tissue,increased NLRP3,caspase-1,ASC,GSDMD protein expression levels in lung tissue,and increased NLRP3,caspase-1,ASC,GSDMD,IL-1 β,IL-18 mRNA ex-pression in lung tissue(P<0.05).Compared with the model group,the pathological injury of lung tissue in the other four groups was improved to varying degrees,the contents of IL-1 β and IL-18 in serum and lung tissue were decreased,the protein expression levels of NLRP3,caspase-1,ASC,GSDMD in lung tissue were decreased,and the mRNA expression levels of NLRP3,caspase-1,ASC,GSDMD,IL-1 β,IL-18 in lung tissue were decreased(P<0.05).Conclusion Qingjin Tongluo Granules may improve lung inflam-mation in MPP mice by regulating NLRP3/Caspase-1/GSDMD mediated cell pyroptosis.
