基于LKB1/AMPK/SIRT1/NF-κB通路探讨款冬花散对支气管扩张症发病机制的研究

目的 探索方药款冬花散通过LKB1/AMPK/SIRT1/NF-κB信号通路对痰热郁肺证支气管扩张症模型大鼠气道炎症和黏液高分泌的影响.方法 将大鼠(70只)随机进行分组,分成7组包括空白组、自身对照组、模型组、罗红霉素组、不同剂量款冬花散组(低、中、高),每组10只大鼠,同时为建立支气管扩张症大鼠模型给予大鼠绿脓杆菌液气管注射.采用款冬花散干预2周后进行取材,观察大鼠肺组织中LKB1、AMPK、SIRT1、NF-κB蛋白和mRNA表达水平.结果 与空白组相比,模型组LKB1、AMPK、SIRT1 mRNA和蛋白表达显著降低,NF-κB mRNA和蛋白表达显著升高(P<0.01).经款冬花散高剂量组治疗后,大鼠肺组织中LKB1、AMPK、SIRT1 mRNA表达水平明显升高,NF-κB mRNA表达水平明显下降,优于款冬花散低剂量组(P<0.01).款冬花散能提高肺组织中LKB1、AMPK、SIRT1蛋白表达水平,降低NF-κB蛋白表达水平,且以款冬花散高剂量组较明显(P<0.01或P<0.05).结论 款冬花散可以通过LKB1/AMPK/SIRT1/NF-κB信号通路改善支气管扩张症痰热郁肺证模型大鼠气道炎症和黏液高分泌状态,并呈一定的量效关系.
Investigation of the Pathogenesis of Bronchiectasis based on the LKB1/AMPK/SIRT1/NF-κB pathway using Kuandonghua Powder
Objective To explore the effect of Kuandonghua Powder on airway inflammation and mucus hypersecretion in the rat model of bronchiectasis with phlegm-heat accumulation in the lungs syndrome through LKB1/AMPK/SIRT1/NF-κB signaling pathway.Meth-ods 70 rats were randomly divided into 7 groups:blank group,control group,model group,roxithromycin group,different doses of Kuandonghua Powder group(low,medium and high),10 rats in each group,and at the same time given the aeruginosa solution to make the rat model of bronchiectasis.The protein and mRNA expression levels of LKB1,AMPK,SIRT1 and NF-κB in rats lung tissue were observed after two weeks of intervention with Kuandonghua Powder.Results Compared with the blank group,the mRNA and protein ex-pression of LKB1,AMPK,and SIRT1 in the model group were significantly reduced,while the mRNA and protein expression of NF-κB were significantly increased(P<0.01).After high-dose treatment with Kuandonghua Powder,the mRNA expression of LKB1,AMPK,SIRT1 were significantly increased,and the expression of NF-κB mRNA was significantly decreased,which was better than the Kuandonghu Powder low-dose group(P<0.01).Kuandonghua Powder can increase the expression of LKB1,AMPK,SIRT1 protein in the lung tissue,and decrease the expression of NF-κB protein,especially in the Kuandonghua Powder high dose group(P<0.01 or P<0.05).Conclusion Kuandonghua Powder can improve the airway inflammation and mucus hypersecretion in the rat model of bronchi-ectasis with phlegm-heat accumulation in the lungs syndrome through the LKB1/AMPK/SIRT1/NF-κB signaling pathway,and it showes a certain dose-effect relationship.
