Investigation of the Pathogenesis of Bronchiectasis based on the LKB1/AMPK/SIRT1/NF-κB pathway using Kuandonghua Powder
Objective To explore the effect of Kuandonghua Powder on airway inflammation and mucus hypersecretion in the rat model of bronchiectasis with phlegm-heat accumulation in the lungs syndrome through LKB1/AMPK/SIRT1/NF-κB signaling pathway.Meth-ods 70 rats were randomly divided into 7 groups:blank group,control group,model group,roxithromycin group,different doses of Kuandonghua Powder group(low,medium and high),10 rats in each group,and at the same time given the aeruginosa solution to make the rat model of bronchiectasis.The protein and mRNA expression levels of LKB1,AMPK,SIRT1 and NF-κB in rats lung tissue were observed after two weeks of intervention with Kuandonghua Powder.Results Compared with the blank group,the mRNA and protein ex-pression of LKB1,AMPK,and SIRT1 in the model group were significantly reduced,while the mRNA and protein expression of NF-κB were significantly increased(P<0.01).After high-dose treatment with Kuandonghua Powder,the mRNA expression of LKB1,AMPK,SIRT1 were significantly increased,and the expression of NF-κB mRNA was significantly decreased,which was better than the Kuandonghu Powder low-dose group(P<0.01).Kuandonghua Powder can increase the expression of LKB1,AMPK,SIRT1 protein in the lung tissue,and decrease the expression of NF-κB protein,especially in the Kuandonghua Powder high dose group(P<0.01 or P<0.05).Conclusion Kuandonghua Powder can improve the airway inflammation and mucus hypersecretion in the rat model of bronchi-ectasis with phlegm-heat accumulation in the lungs syndrome through the LKB1/AMPK/SIRT1/NF-κB signaling pathway,and it showes a certain dose-effect relationship.
