The effects of Zhenwu Tang and Yuemaitang on the expression of t-PA/PAI-1 in renal tis-sue of rats with doxorubicin induced nephropathy
Objective Observing the effects and differences of Zhongjingli Water Formula Zhenwu Tang and Yuenu Tang on the expres-sion of tissue type plasminogen activator(t-PA)and plasminogen activator inhibitor-1(PAI-1)in renal tissue of rats with doxorubi-cin nephropathy model.Methods 48 male SD rats were randomly divided into a normal group of 12,a modeling group of 36,and a modeling group of 2 tail vein injections of doxorubicin(4mg·kg-1,2mg·kg-1)for modeling.After successful modeling,the rats were randomly divided into a model group,a Zhenwu Tang group,and a Yuenu Tang group,with 12 rats in each group.The corresponding drugs were administered orally for 6 weeks.Detect 24-hour total urine protein(24h-UTP),serum albumin(ALB),serum creatinine(Scr),and urea nitrogen(BUN)in rats;Observation of pathological morphological changes in rat kidneys under light microscopy;Im-munohistochemical detection of t-PA and PAI-1 expression in rat kidney tissue.Results Compared with the normal group,the patho-logical damage to the kidneys of the model group rats was significant,with glomerular hypertrophy,tubular dilation,extracellular matrix proliferation,and a large number of inflammatory cell infiltration in the stroma;24h-UTP,serum Scr,BUN,renal tissue PAI-1 ex-pression increased(P<0.05),while ALB and renal tissue t-PA expression decreased(P<0.05);Compared with the model group,the renal pathological damage of rats in the Zhenwu Tang and Yuemaitang groups was significantly improved,with decreased expression of 24h-UTP,serum Scr and renal tissue PAI-1(P<0.05),and increased expression of ALB and t-PA in renal tissue(P<0.05).Conclusion Zhenwu Tang and Yuemaitang may improve renal injury in rats with doxorubicin nephropathy by affecting the expression of t-PA/PAI-1 in renal tissue.
