基于miRNA-21a-5p/STAT3轴探讨五虎汤对呼吸道合胞病毒诱导的外泌体诱发哮喘的影响

目的 基于miRNA-21a-5p/STAT3轴探讨五虎汤对呼吸道合胞病毒(RSV)诱导的外泌体诱发哮喘的影响.方法 SPF级C57小鼠40只,随机分为空白组、模型组、五虎汤组和利巴韦林组,空白组给予PBS滴鼻,其余组给予RSV感染的骨髓间充质干细胞外泌体(BMSCs-Exo-RSV)PBS稀释后滴鼻,隔天1次,共7次.干预结束后24h,空白组、模型组给予蒸馏水灌胃,五虎汤组给予五虎汤煎剂灌胃,利巴韦林组给予利巴韦林稀释水溶液灌胃,共干预1周.干预期间观察各组小鼠的一般行为变化,评估有无哮喘症状和体征的出现,干预结束后取小鼠肺组织检测.使用HE、Masson染色观察肺组织炎症浸润和胶原沉积情况,RT-qPCR法检测肺组织中miRNA-21a-5p及STAT3 mRNA表达情况,Western blot法检测免疫组化法检测肺组织中STAT3、p-STAT3蛋白表达情况,体外双荧光素酶实验验证miRNA-21与STAT3 mRNA的靶向调控作用.结果 与空白组相比,模型组小鼠出现哮喘典型改变,肺组织中炎症浸润和胶原沉积明显增多,肺组织中miRNA-21a-5p、STAT3 mRNA和p-STAT3表达水平升高(P<0.01).与模型组相比,五虎汤组小鼠症状和体征改善,肺组织中miRNA-21a-5p、STAT3 mRNA和p-STAT3表达水平均下降(P<0.01).体外双荧光素酶结果显示:miRNA-21a-5p能够靶向促进STAT3 mRNA的表达(P<0.05).结论 BMSCs-Exo-RSV能够激活miRNA-21a-5p/STAT3轴,促进小鼠肺组织中miRNA-21a-5p、STAT3mRNA和p-STAT3蛋白的表达,引起气道炎症和重塑,从而诱发哮喘,而五虎汤则能通过逆转以上环节改善哮喘.
Based on miRNA-21a-5p/STAT3 axis,this paper discusses the effect of WuHuTang on asthma triggered by exosomes induced by RSV
Objective Based on the miRNA-21a-5p/STAT3 axis,to explore the effect of WuHuTang on asthma triggered by the exo-somes induced by RSV.Methods Forty SPF C57 mice were randomly divided into blank group,model group,WuHuTang group and ribavirin group.The blank group was given PBS nasal drops,and the rest groups were given PBS diluted with RSV-infected bone mar-row mesenchymal stem cell exosomes(BMSCs-EXO-RSV)nasal drops,once every other day for 7 times.Twenty-four hours after the intervention,the blank group and the model group were given distilled water by gavage,the WuHuTang group was given WuHuTang by gavage,and the ribavirin group was given ribavirin diluted aqueous solution by gavage for one week.During the intervention,the gen-eral behavior changes of mice in each group were observed,and the symptoms and signs of asthma were evaluated.After the intervention,the lung tissue of mice was taken for detection.HE and Masson staining were used to observe the inflammatory infiltration and collagen deposition in lung tissue,RT-QCPR method was used to detect the expression of miRNA-21 a-5p and STAT3 mRNA in lung tissue,Western blot method was used to detect the expression of STAT3 and p-STAT3 protein in lung tissue,and double luciferase experiment in vitro verified the targeted regulation of miRNA-21 and STAT3 mRNA.Compared with the blank group,the model group showed typ-ical changes of asthma,inflammatory infiltration and collagen deposition in lung tissue increased significantly,and the expression levels of miRNA-21a-5p,STAT3 mRNA and p-STAT3 in lung tissue increased(P<0.01).Compared with the model group,the symp-toms and signs of WuhuTang group were improved,and the expression levels of miRNA-21a-5p,STAT3 mRNA and p-STAT3 in lung tissue were decreased(P<0.01).The results of double luciferase in vitro showed that miRNA-21a-5p could promote the ex-pression of STAT3 mRNA(P<0.05).Conclusion BMSCs-Exo-RSV can activate the miRNA-21a-5p/STAT3 axis,promote the expression of miRNA-21a-5p,STAT3mRNA andp-STAT3 protein in mouse lung tissue,cause airway inflammation and remodeling,and thus induce asthma,while WuHuTang can improve asthma by reversing the above links.
