九蒸九晒多花黄精的多个成分动态研究

目的 采用紫外可见-分光光度法与高效液相色谱法,分析九蒸九晒多花黄精的多个成分含量变化趋势。方法 采用Agilent ZORBAX-Extend-C18(250 mm×4。6 mm,5 μm)色谱柱,以甲醇-水为流动相,测定5-羟甲基糠醛、5-羟基麦芽酚、黄精碱A的含量;采用色谱柱Prevail carbohydrate ES(4。6 mm ×250 mm,5 μm),以乙腈-水为流动相测定果糖、蔗糖、葡萄糖的含量;采用UV-2450紫外-可见分光光度计测定多糖与皂苷的含量;参照2020版《中国药典》测定醇溶性浸出物。分析各成分随蒸晒次数增加的含量变化。结果 随着多花黄精蒸晒次数的增加,5-羟甲基糠醛、5-羟基麦芽酚与黄精碱A的含量会整体增加,多糖、皂苷、葡萄糖与醇溶性浸出物的含量会整体下降,蔗糖的含量先增加后下降。结论 蒸晒次数对于各成分的含量影响很大,可以通过控制九蒸九晒的炮制工艺,调节多花黄精的各成分含量,为其炮制过程的质量控制提供理论依据。
Dynamic study on multiple components of Polygonatum cyrtonema
Objective Ultraviolet-visible spectrophotometry and High performance liquid chromatography(HPLC)were used to analyze the content change trend of multiple components in Polygonatum cyrtonema.Methods Agilent ZORBAX-Extend-C18(250 mm × 4.6 mm,5 μm)column was used with methanol-water as mobile phase for the determination of 5-Hydroxymethylfurfural,5-Hydroxy-maltol and polygonatine A.The content of fructose,glucose and sucrose were determined by chromatographic column Prevail carbohydrate ES(4.6 mm × 250 mm,5 μm)with acetonitrile-water as mobile phase.The contents of polysaccharides and saponins were deter-mined by UV-2450 UV-visible spectrophotometer.The alcohol-soluble extract was determined according to the 2020 edition of Chi-nese Pharmacopoeia'.The content changes of each component with the increase of steaming times were analyzed.Results With the in-crease of steaming times of Polygonatum cyrtonema,the contents of 5-Hydroxymethylfurfural,5-Hydroxymaltol and polygonatine A in-creased,the contents of polysaccharides,saponins,glucose and alcohol-soluble extract decreased,and the content of sucrose increased first and then decreased.Conclusion The number of steaming and drying times has a great influence on the content of each component.The content of each component of Polygonatum cyrtonema can be controlled by the processing technology of nine-steam-nine-bask,which provides a theoretical basis for the quality control of its processing process.
