Zhuang-Gu-Fang regulates the balance of bone and lipid formation in senile osteoporosis mice by Foxp1 and PPARγ
Objective To study Zhuang-Gu-Fang regulates the balance of bone and lipid formation in senile osteoporosis mice by Foxp1 and PPARy.Methods 30 SAMP6 mice were randomly divided into model group,calcium carbonate+alfaD3 group and Zhuang-gu-fang group,with 10 mice in each group.10 SAMR1 mice were selected as blank group.After 8 weeks of continuous intervention,changes in bone microstructure were detected by Micro CT,and osteogenic factors OCN,Osterix,DMP1,lipogenic factors FABP4,and Cebpa were detected by IF staining.The protein and mRNA expression levels of Foxp1,Notch1 and PPARy were analyzed by Western blot and RT-PCR.Results Compared with blank group,BV/TV,Tb.Th and Tb.N in model group showed a significant decrease(P<0.05),Tb.Sp and SMI were significantly increased(P<0.01),while the levels of OCN,Osterix and DMP1 in bone tissue were signifi-cantly decreased(P<0.01).The levels of FABP4 and Cebpa were significantly increased(P<0.01),while the mRNA and protein ex-pression levels of Foxp1 and Notch1 were significantly decreased(P<0.01),the mRNA and protein expression levels of PPARγ were significantly increased(P<0.01).Effect of Zhuang-Gu-Fang on BV/TV、Tb.Th、Tb.N significantly increased(P<0.05),while Tb.Sp and SMI were significantly decreased(P<0.01).The mRNA and protein levels of OCN,Osterix,DMP1 and Foxp1,Notch1 were increased in bone tissue(P<0.05),while the mRNA and protein levels of FABP4,Cebpa and PPARγ were significantly de-creased(P<0.01).Conclusion Zhuang-Gu-Fang may improve the balance of bone and lipid formation by increasing Foxp1 and Notch1 in bone tissue and decreasing PPARy level.
