祛痰化瘀方对AF大鼠心房代谢重构的机制研究

目的 研究祛痰化瘀方对AF大鼠心房代谢重构的作用机制。方法 雄性SD大鼠120只,随机分为正常组、模型组、祛痰化瘀方低、中、高剂量组,盐酸胺碘酮组,使用Ach-CaCl2尾静脉注射诱导AF模型;造模成功后,给予祛痰化瘀方低、中、高剂量及盐酸胺碘酮治疗,治疗后检测各组大鼠心电图、AF持续时间、FFA浓度、TG浓度,qPCR法检测各组大鼠心房腺苷酸激活蛋白激酶(AMP-activated protein kinase,AMPK)、乙酰辅酶 A 羧化酶(acetyl CoA carboxylase,ACC)、CPT-1B 肉碱脂酰转移酶(camitine palmitoyltransferase-1B,CPT-1B)、脂肪酸转位酶(fatty acid translocase,FAT/CD36)的 mRNA 表达,Western blot法检测各组大鼠心房AMPK、p-AMPK、ACC、p-ACC、CPT-1B的蛋白表达,油红O染色观察各组大鼠心房脂质沉积情况。结果 祛痰化瘀方及盐酸胺碘酮治疗后,AF持续时间缩短(P<0。05),心房中FFA、TG浓度下降(P<0。05),AMPK、ACC、CPT-1B、FAT/CD36 的 mRNA 表达升高(P<0。05),p-AMPK/AMPK、p-ACC/ACC、CPT-1B 的相对蛋白表达升高(P<0。05),心房中脂质沉积情况改善,脂滴数量减少。结论 祛痰化瘀方能够明显改善AF大鼠的心房代谢重构,其作用机制可能与激活AMPK/ACC/CPT-1B通路有关。
Mechanism of QuTan Hua Yu Fang on metabolic remodeling of atria in AF rats
Objective To study the mechanism of QuTan HuaYu Fang on atrial metabolic remodeling in AF rats.Methods 120 male SD rats were randomly divided into normal group,model group,QuTan HuaYu Fang low,medium and high dose group,and amiodarone hydrochloride group,and the AF model was induced by caudal vein injection of Ach-CaCl2;after the modeling was successfully estab-lished,QuTan HuaYu Fang was given low,medium and high doses and amiodarone hydrochloride treatment;after treatment,electrocardi-ogram,AF duration,FFA concentration and TG concentration of rats in each group were tested,the mRNA expression of AMPK,ACC,CPT-1B,and FAT/CD36 was detected by qPCR.The protein expression of AMPK,p-AMPK,ACC,p-ACC,and CPT-1B were detected in the atria of rats in each group by Western blot method,and the lipid deposition in the atria of rats in each group was observed by oil red O staining.Results The duration of AF was shorter after treatment with QuTan HuaYu Fang and amiodarone hydrochloride(P<0.05),and the concentrations of FFA and TG in the atria were decreased after treatment with QuTan HuaYu Fang(P<0.05),and the concentrations of AMPK,ACC,and CPT-1B,FAT/CD36 mRNA expression increased(P<0.05),the relative protein expression of p-AMPK/AMPK,p-ACC/ACC,CPT-1B increased(P<0.05),lipid deposition in the atria improved,and the number of lipid droplets decreased.Conclusion QuTan HuaYu Fang can significantly improve atrial metabolic remodeling and intervene in AF rats,and its mech-anism of action may be related to the activation of AMPK/ACC/CPT-1B pathway.
