Exploring the Regulation of Autophagy in Granulosa Cells of PCOS Phlegm-Dampness Syn-drome Model Rats by Qi Gong Wan through the AMH/AMHR Ⅱ Pathway
Objective This study aimed to investigate the effects of Qi Gong Wan on the expression of AMH/AMHR Ⅱ in ovarian granu-losa cells and autophagy in rats with polycystic ovary syndrome(PCOS)and phlegm-dampness syndrome.Methods A PCOS model was established and groups were divided.HE staining,ELISA,immunohistochemistry,and Western Blot were used to detect ovarian pa-thology,serum hormone levels,LC3 distribution,and vascular changes.Results The levels of LH,T,and Ins in the serum of the model group were significantly increased,while the Chinese medicine group could significantly reduce these hormone levels.Pathological sec-tions showed that the Chinese medicine group improved ovarian morphology and reduced the number of immature follicles.Immunohisto-chemical and Western blot analyses indicated that the Chinese medicine group could downregulate the expression of LC3,AMH,AMHR Ⅱ,Beclin-1,and LC3A/B,upregulate the expression of p62,and inhibit excessive autophagy.Conclusion Qi Gong Wan may im-prove ovarian function in PCOS rats by regulating the AMH/AMHR Ⅱ signaling pathway.
