Effects of He-Mu Adjunct Electroacupuncture points on Heart Rate Variability and Serum cAMP,cGMP and SP in Irritable Bowel Syndrome Rats
Objective To compare the effects of"ST37"and"ST25"on heart rate variability and serum cAMP,cGMP and SP levels in rats with IBS model.We investigated whether there was a synergistic effect of the combined acupoints in regulating the autonomic balance of IBS rats.Methods 55 male clean-grade Wistar rats were selected and randomly divided into blank group(10 rats)and modelling group(45 rats),and the modelling group was prepared by acute and chronic combined IBS model,and the successfully prepared IBS model rats were randomly divided into the modelling group(10 rats),the single-point group(10 rats),and the combined-point group(10 rats);the single-point group and the combined-point group were continuously intervened by electroacupuncture for 14 days;after the electroacupuncture treatment,the rats in each group were recorded.After treatment,the body weight,semi-quantitative AWR score,behavioural and HRV measurements,and serum cAMP,cGMP and SP levels were recorded in each group.Results Before inter-vention,compared with the blank group,rats in the model group and the electroacupuncture treatment group had lower body mass,higher AWR scores,and increased visceral sensitivity(P<0.05,P<0.01);after treatment,compared with the model group,electroacupunc-ture significantly increased body mass(P<0.01),decreased visceral sensitivity(P<0.05,P<0.01),increased the number of times the rats entered the open arm,the residence time,and total distance travelled by the activity trajectory(P<0.05,P<0.01),decreased LF/HF(P<0.05)and elevated HF as well as RMSSD(P<0.05),and decreased serum cAMP、cGMP and SP content(P<0.01,P<0.05,P<0.01);there were no statistical differences in body mass,AWR score,behavioural,HRV,and serum cAMP,cGMP and SP content of the rats in the electroacupuncture single-point group compared with the combination and matching group.Conclusion Both the application of single acupoint and combined acupoints have been demonstrated to effectively enhance visceral sensitivity in IBS pa-tients,alleviate anxiety and negative affect,modulate heart rate variability,and decrease the serum levels of cAMP,cGMP,and sub-stance P.but the use of combined acupoints did not exhibit an additive synergistic effect.
