Based on data mining to explore the law of traditional Chinese medicine for the decline of o-varian reserve
Objective Exploring the medication pattern of Professor Wu Xianqun in treating ovarian reserve dysfunction(DOR)and ver-ifying it through animal experiments.Methods Building a database based on the ancient and modern medical case cloud platform,col-lecting DOR cases treated by Wu Xianqun from 2019 to 2021,analyzing medication patterns,and screening basic prescriptions.On this basis,DOR model rats were given basic formula intervention,and the ovulation cycle of the model rats was observed through vaginal smears;Evaluation of ovarian function using eosin hematoxylin(HE)staining;Enzyme linked immunosorbent assay(ELISA)was used to detect serum levels of anti Maitreya hormone(AMH)and estradiol(E2);Western blot(WB)was used to detect the protein levels of hypoxia inducible factor 1 alpha(HIF-1 alpha)and vascular endothelial growth factor A antibody(VEGF).Results There are a total of 49 traditional Chinese medicines used to treat DOR,with Angelica sinensis having the highest frequency of use at 138 times;The high-est frequency of effectiveness is in nourishing blood and promoting blood circulation;Analysis of association rules for drug combinations:licorice Angelica sinensis,Angelica sinensis licorice,and yam Angelica sinensis are common;The core drugs of the basic formula consist of yam,Cornus officinalis,stir fried Atractylodes macrocephala,Rehmannia glutinosa,Goji berries,Chuanxiong,Angelica sinensis,Paeonia lactiflora,licorice,and Aconitum carmichaelii.Animal experiments have confirmed that compared with the blank group,the model group rats have disrupted estrous cycles,significantly reduced the number of follicles at all levels,significantly increased serum E2,significant-ly decreased AMH,and significantly increased the expression of HIF-1α and VEGF in the ovaries(P<0.01);Compared with the model group,the high-dose group of traditional Chinese medicine basic formula showed a reversal of the above indicators(P<0.01).Conclusion The core drug used by Professor Wu Xianqun to treat DOR is indeed effective,and its mechanism may be to increase follicle production through the HIF-1α/VEGF signaling pathway.
