Effect of Curcumin on Proliferation and Migration of Lung Adenocarcinoma Cells by Inhibiting the Secretion of Microvesicles
Objective To investigate the effect of curcumin on the proliferation and migration of lung adenocarcinoma A549 cell line and its possible mechanism.Methods A549 cell lines were treated with different concentrations of curcumin,and supernatant microvesicles were extracted,identified and counted by flow cytometry.A549 cells were further treated with curcumin and curcumin+microvesicles,and cell proliferation activity was detected by CCK8 assay.Cell migration function was detected by scratch assay and Transwell assay.Results Flow cytometry analysis showed that as the concentration of curcumin increased,the number of microvesicles secreted by A549 cells significantly decreased;After adding curcumin or curcumin+microvesicles treatment to A549 cell line,the results of CCK8 cell proliferation experiment showed that curcumin could inhibit A549 cell proliferation,microvesicles could reverse the inhibitory effect of curcumin,scratch test and Transwell test results showed that curcumin could inhibit A549 cell migration,microvesicles could reverse the inhibitory effect of curcumin,and the difference between groups was statistically significant(P<0.05).Conclusion Curcumin can reduce the secretion of microvesicles in A549 cell line,thereby inhibiting the proliferation and migration function of lung adenocarcinoma.
