摘要
为研究养心安眠胶囊治疗心气虚的药理作用和量效关系.本研究采用"惊劳、饥饿结合损耗心气"方法,建立小鼠心气虚模型,设计空白组、模型组、阳性药组、养心安眠胶囊0.273 g/kg组、0.546 g/kg组、0.819 g/kg 3个给药组,通过检测小鼠游泳力竭时间、肿瘤坏死因子-α(TNF-α),心肌组织匀浆中超氧化物歧化酶(SOD)、丙二醛(MDA)、肌酸激酶(CK)、乳酸脱氢酶(LDH)等指标,研究养心安眠胶囊治疗心气虚的药理作用和量效关系.结果表明,与模型组相比,养心安眠胶囊0.273 g/kg组对小鼠游泳力竭时间、TNF-α、SOD、MDA无显著性影响(P＞0.05),但可显著降低CK、LDH水平(P＜0.05);养心安眠胶囊0.546 g/kg组和0.819 g/kg组对各指标均有显著性影响(P＜0.01),表现为明显延长小鼠游泳力竭时间、明显降低TNF-α、MDA、CK、LDH水平,显著提高SOD水平.表明养心安眠胶囊具有显著的治疗心气虚的药理作用,养心安眠胶囊给药剂量在0.273 g/kg～0.546g/kg范围内,有良好的量效相关性,在0.546 g/kg和0.819 g/kg给药剂量下药效作用明显.本研究为后期养心安眠胶囊临床用药提供理论参考.
Abstract
In order aims to study the pharmacological action and dose-effect relationship of Yangxin Anmian capsule in the treatment of heart-qi deficiency.This study establisbed the model of Heart Qi deficiency in mice by the method of"fatigue,hunger combined with the loss of mental energy".The experiment set up blank group,model group,positive drug group,Yangxin Anmian capsule 0.273 g/kg group,0.546 g/kg group,and 0.819 g/kg group.The exhaustive swimming time,tumor necrosis factor(TNF-α),superoxide dismutase(SOD),malondialde-hyde(MDA),creatine kinase(CK)and lactate dehydrogenase(LDH)were measured,to investigate the pharmacological action and dose-effect relationship of Yangxin Anmian capsule in the treatment of heart-qi deficiency.The results showed that compared with the model group,Yangxinanmian capsule 0.273 g/kg group had no significant effect on the exhaustive swimming time,TNF-α,SOD and MDA(P＞0.05),but significantly decreased the levels of CK and LDH(P＜0.05).Yangxinanmian capsule 0.546 g/kg group and 0.819 g/kg group had sig-nificant effect on all indexes(P＜0.05 or P＜0.01),the results showed that the time of swimming exhaustion was prolonged,the levels of TNF-α,MDA,CK and LDH were decreased,and the levels of SOD were increased.Yangxinanmian capsule has a significant pharmacolog-ical effect in treating heart-qi deficiency,and there is a good dose-effect correlation in the range of 0.273 g/kg～0.546 g/kg,and the effect was obvious at the dosage of 0.546 g/kg and 0.819 g/kg.This study provides theoretical reference for the later clinical medication of Yangxin Anmian capsule.
