基于UPLC-Q-Exactive-MS代谢组学技术的蒺藜果提取物对缺血性脑中风大鼠的保护作用及机制研究

采用LC-MS为基础的代谢组学技术,研究蒺藜果提取物对大脑中动脉阻塞(MCAO)致缺血性脑中风(IS)大鼠的保护作用.30只大鼠随机分为假手术组、模型组和给药组,以大鼠神经症状评分和脑缺血面积百分比为指标,评价蒺藜果提取物对IS大鼠的保护作用.基于LC-MS的非靶向代谢组学的方法对大鼠尿液进行研究,获取蒺藜果提取物可调控IS的差异代谢物,并对其进行代谢通路分析.蒺藜果提取物能显著降低MCAO大鼠的神经症状评分和脑梗死面积;通过分析模型组和假手术组尿液代谢谱,筛选并鉴定出差异代谢物20个,主要涉及3条代谢通路.蒺藜果提取物主要通过调控缬氨酸、亮氨酸及异亮氨酸的生物合成、三羧酸酸循环(TCA循环)和叶酸生物合成起到抗IS的作用,为临床应用提供理论依据.
The Protective Effect and Mechanism of Tribulus terrestris L.Fruit Extract on Ischemic Stroke Rats Based on UPLC-Q-Exactive-MS Metabolomics
To investigate the protection effect of Tribulus terrestris L.fruit extract on the middle cerebral artery occlusion(MCAO)-induced IS in rats based on the LC-MS metabolomics technique.30 rats were divided into the sham group randomly,the model group and adminis-tration group.The anti-IS effect of T terrestris fruit extract was conducted by the evaluation of neurological defects and cerebral infarct vol-ume percentage in rats.Based on the LC-MS untargeted metabolomics approach was used to conduct on rat urine to obtain the differential metabolites of T terrestris fruit extract anti-IS and analyze their metabolic pathways.T terrestris fruit extract treatment significantly lowered neurological deficit scores and cerebral infarct volume percentage in MCAO rats.By analyzing the urine metabolic profiles of model group and sham group,20 kinds of the differential metabolites were screened and identified,mainly involving three metabolic pathways.T terre-stris fruit extract is mainly through regulating the metabolism of valine,leucine and isoleucine biosynthesis,citrate cycle(TCA cycle)and folate biosynthesis to protect IS rats,so as to provide a theoretical basis for the rational clinical application.
