Study on Sampling and Preservation Methods for Morphological Identification of Honeybee Resources
In order to improve the efficiency of bee morphological identification,morphological identification is an essential technique for bee germplasm evaluation.In this study,different sampling and preservation methods were used to compare the kissing rate,the amount of tissue damage,the influence of chroma and the influence of toughness of bee samples in different experimental groups.The results showed that there was no significant difference in the kissing rate of B fumigation at 20 s and 30 s in experimental groups(P>0.05),but it was sig-nificantly higher than that in other experimental groups(P<0.05).The number of honeybee tissue damage in experimental group D was significantly lower than that in experimental group E,F,G and control group(P<0.05).Storage at 4 ℃ for 1 to 3 months had no effect on the chroma and toughness of honeybee sampls.It is recommended to use CO2 at the flow rate of 2 L/min for 20 s,soak in the preservation solution of 75%alcohol and 0.5%glycerol,and complete the morphological identification within 3 months.
