Exploring the Mechanism of Action of Helicirysum arenarium Alcoholic Extracts in the Treatment of Nonalcoholic Steatohepatitis based on in Vitro Experiments and Network Pharmacology
The pharmacodynamic basis and predicted mechanism of action of Helicirysum arenarium alcoholic extracts for the treatment of non-alcoholic steatohepatitis were investigated based on in vitro experiments and network pharmacology method.an in vitro NASH model was established with free fatty acid induced HepG2 cells and intervened with different concentrations of Helicirysum arenarium alcoholic extract,which was found to have a good ameliorative effect on lipid deposition in the NASH cell model;in vitro oil adsorption experiments and experiments on lipase activity of alcoholic extracts of Helicirysum arenarium were conducted,and it was found that Helicirysum arena-rium has better oil adsorption ability and the ability to inhibit lipase activity,indicating that Helicirysum arenarium has good lipid-lowering activity in vitro.On the basis of the above,CNKI,Wanfang database,Pubmed and other databases were utilized to find saxifrage-related tar-gets,and Gene Cards,OMIM,TTD,Drug Bank platforms were used to find non-alcoholic steatohepatitis-related proteins,,and 33 potential active components and 88 NASH-related targets were obtained,and the core targets were obtained through protein-protein interactions analysis,and the top 10 nodes were selected.The mechanism of action may be related to AGE-RAGE signaling pathway,lipid and athero-sclerosis pathway,insulin resistance pathway,chemo-carcinogenic-multi-reactive oxygen species pathway,adipocytokine signaling pathway and HIF-1 signaling pathway in diabetic complications,etc.through a multi-pathway and multi-target approach for the treatment of NASH.The molecular docking of core targets and core components was performed using AutoDockTools 1.5.7 software,and the results showed that both had good binding ability.In conclusion,this study initially explored the"active ingredient-target-pathway"mechanism of action of Helicirysum arenarium alcoholic extracts for the treatment of NASH,which provides a reference for the validation of its mechanism of action and also provides a basic basis for the possible use of Helicirysum arenarium as a treatment for NASH.
