Antipyretic and Antitussive Effects of Chaixing Granules on Lung the Effect of MPO,NO and iNOS Expression
To observe the antipyretic and anticough effects of Chaixing granules on the levels of myeloperoxidase(MPO),nitric oxide(NO)and inducable nitric oxide synthase(iNOS)protein expression in lung.50 screened ICR mice were randomly divided into normal group,Xiaoer Chaigui Tuire granule group(5.2 g/kg),Chaixing granule high-dose,medium-dose and low-dose groups(3 g/kg,1.5 g/kg,0.75 g/kg),normal group was given distilled water,and other groups were given corresponding liquid for 3 consecutive days.Once a day,after the last dose,the mice were put into a beaker of concentrated ammonia water to induce cough,and the cough incubation period and cough times of the mice within 3 minutes were recorded.Sixty qualified SD rats were randomly divided into normal group,model group,aspirin enteric co-ated tablet group(0.1 g/kg),Chaixing granule high-dose,medium-dose and low-dose groups(2.1 g/kg,1.05 g/kg,0.525 g/kg).Normal group and model group were given distilled water,and other groups were given corresponding liquid by intragastric administration,once a day.For three consecutive days.After the last administration,the normal group was injected with normal saline subcutaneously,and the other groups were injected with dry yeast suspension subcutaneously.The temperature changes of each group were detected,the rats were killed,MPO activity and NO level in lung were determined,iNOS protein expression was detected by immunohistochemistry.Compared with the normal group,each dose group of Chaixing granule had better antitussive effect(P<0.01),and the low dose group had better antipyretic ef-fect(P<0.01 or P<0.001).Compared with normal group,the MPO activity in model group was significantly increased(P<0.05),the ex-pression ofiNOS and NO levels were significantly increased(P<0.001),and the activities of MPO and NO were decreased in high and low dose Chaixing granule groups(P<0.05 or P<0.01).iNOS expression in alveolar epithelial cells was significantly decreased in each dose group of Chaixing granules(P<0.001).Chaixing granules have good antitussive effect,the mechanism of antitussive action is related to the decrease of MPO and NO activity and the expression of iNOS in lung.
