Isolation and Identification of a Muscovy Goose Parvovivus in Guangxi Region
In order to reveal the molecular epidemiological characteristics of a goose parvovivus from muscovy in Guangxi region and its genetic variation mechanism.In this study,PCR was used to identify the pathogen of the disease materials sent for inspection by a muscovy duck farm;the identified positive samples were inoculated into allantoic cavity of 12~13 day-old muscovy duck embryos to isolate the virus.The gene was amplified,cloned and sequenced;and the gene sequences of reference strains were compared and analyzed to construct a phy-logenetic tree.The results showed that Muscovy duck-origin goose parvovirus(MDGPV)was isolated successfully and named as GXBH strain;the homology between this isolation and 17 isolates of waterfowl parvovirus in GenBank was 79.4%~98.7%;The isolated strain had higher homology with the representative strains of GPV,while it had lower homology with the representative strains of MDPV.The gene shared 93.3%~98.7%nucleotide homology to Goose parvovirus(GPV)strains and 79.4%~79.8%nucleotide homology to Muscovy duck parvorirus(MDPV).It was in a different branch of the evolutionary tree with the MDPV strains,and was in the same branch with the GPV strains.The results suggested that the isolated strain was MDGPV,which had similar genetic evolutionary relationships with the represen-tative strains of GPV.
