为培养龙岩山麻鸭鸭胚成纤维细胞。本研究选择龙岩山麻鸭10日龄鸭胚组织,采用组织块法和酶消化法进行成纤维细胞培养,利用免疫荧光法对培养的成纤维细胞进行鉴定。结果表明,培养的鸭胚成纤维细胞12h后呈典型的长梭形,48h后,细胞轮廓清晰,核仁明显,传至第3代的成纤维细胞经DAPI和绿色荧光标记染色后均呈阳性,具有成纤维细胞的特征。本研究成功从龙岩山麻鸭鸭胚组织中分离培养出成纤维细胞,为后期龙岩山麻鸭开展分子生物学和细胞生物学等研究提供良好的基础。
Culture and Identification of Embryo Fibroblasts from the Longyan Shan-Ma Duck
To cultivate the duck embryo fibroblasts from Longyan Shan-Ma duck.The 10-day-old duck embryo tissues of Longyan Shan-Ma duck were used for the fibroblast culture by the tissue block method and enzyme digestion method,and the cultured fibroblasts were identified by the immunofluorescence method.The results indicated that the cultured duck embryo fibroblasts showed a typical long spindle shape after 12 h,and the outline of the cells were well-defined and the nucleoli were distinct after 48 h.The 3rd generation fibroblasts were all positive after DAPI and green fluorescence staining with the characteristic of fibroblasts.Fibroblasts were successfully isolated and cul-tured from the embryo tissue of the Longyan Shan-Ma duck,which provides good materials for the late research of molecular biology and cell biology of the Longyan Shan-Ma duck.
