Isolation,Culture and Identification of Liver Mesenchymal Stem Cells in Longyan ShanMa Duck
In order to explore the in vitro cell preservation method of Longyan Shanma duck,this study established an in vitro culture system of embryonic liver mesenchymal stem cells,so as to achieve the purpose of using the cells to protect the excellent species resources.Liver mesenchymal stem cells(LMSCs)were isolated by trypsin digestion from 18~20 days old healthy Longyan Shanma duck embryos and cul-tured in vitro for cell morphology observation.CD29 and CD44 surface markers were detected by immunofluorescence,and the positive rate was analyzed.LMSCs from Longyan Shanma duck embryos proliferated well in vitro,and LMSCs positively expressed CD29 and CD44,with positive expression rates of 95.20%and 93.46%,respectively.LMSCs were isolated and cultured in vitro from embryonic liver of Longyan Shanma duck.It can provide research ideas for waterfowl stem cell research,and provide reference for the conservation and utilization of germplasm resources of Longyan Shanma duck.
