为探索龙岩山麻鸭体外细胞保存方式,本研究建立了胚胎肝间充质干细胞的体外培养体系,从而达到利用细胞保护优异物种资源的目的.选用18~20日龄健康龙岩山麻鸭鸭胚通过胰蛋白酶消化分离出肝间充质干细胞(LMSCs),进行体外培养,细胞形态观察.通过免疫荧光检测CD29和CD44表面标记物,并进行阳性率分析.龙岩山麻鸭胚胎LMSCs体外增殖能力良好.LMSCs阳性表达了 CD29和CD44,阳性表达率分别为95.20%和93.46%.体外分离培养出龙岩山麻鸭胚胎LMSCs,可为水禽干细胞研究提供研究思路,并为龙岩山麻鸭种质资源保护和利用提供参考.
Isolation,Culture and Identification of Liver Mesenchymal Stem Cells in Longyan ShanMa Duck
In order to explore the in vitro cell preservation method of Longyan Shanma duck,this study established an in vitro culture system of embryonic liver mesenchymal stem cells,so as to achieve the purpose of using the cells to protect the excellent species resources.Liver mesenchymal stem cells(LMSCs)were isolated by trypsin digestion from 18~20 days old healthy Longyan Shanma duck embryos and cul-tured in vitro for cell morphology observation.CD29 and CD44 surface markers were detected by immunofluorescence,and the positive rate was analyzed.LMSCs from Longyan Shanma duck embryos proliferated well in vitro,and LMSCs positively expressed CD29 and CD44,with positive expression rates of 95.20%and 93.46%,respectively.LMSCs were isolated and cultured in vitro from embryonic liver of Longyan Shanma duck.It can provide research ideas for waterfowl stem cell research,and provide reference for the conservation and utilization of germplasm resources of Longyan Shanma duck.
