Application of Auxin Analogues in Tissue Culture of Polyploid Dandelion
The aim of this paper is to study the effects of different auxin species and concentrations on tissue culture of dandelion,and provide reference for the rapid utilization of polyploid dandelion.In this study,the effects of different auxin types and concentrations on polyploid dandelion tissue culture were studied by using colchicin-induced doubled medicinal dandelion leaves as materials.The results showed that the optimal medium for callus induction was MS+2,4-D(0.1 mg/L)+NAA(0.5 mg/L)+6-BA(1.5 mg/L).Callus induction rate was 100%on 6 days after inoculation.Callus were moist and dense,green and shiny,and a large number of advective buds were spontaneously generated,with an average number of 10.66 buds.The optimal medium for proliferation and differentiation was MS+NAA(0.5 mg/L)+6-BA(1.5 mg/L)+2,4-D(0.1 mg/L),the differentiation rate reached 97.74%and the multiplication ratio was 10.8.The optimal rooting medium was 1/2MS+IBA(1.5 mg/L)+NAA(1mg/L)+activated carbon(0.8 g/L),and the roots were robust,the number of roots was 11.34,and the rooting fre-quency was up to 93.47%.The survival rate of transplanting plants reached 100%.The effects of different auxin and auxin concentration on tissue culture of dandelion are obviously different.The concentration of auxin should be adjusted in time according to the variety and absor-ption characteristics of experimental materials to improve the survival rate and utilization rate of dandelion.This study provides experimental support for the establishment of polyploid dandelion tissue culture system and lays the foundation for further deepening research.
