Protective Effect of Pinolenic Acid in Pinus Koraiensis Seed Oil on Human Coronary Artery Endothelial Cells Injury Based on Pinolenic Acid Targets HMGCR,a Key Enzyme in Lipid Synthesis
Pinus koraiensis seed oil extracted from red pine seed is a natural vegetable oil,which contains a special unsaturated fatty acid-pinolenic acid.In this study,pinolenic acid,an active ingredient Pinus koraiensis seed oil,was used to target HMGCR,a key enzyme in lipid synthesis,to protect human coronary artery endothelial cells and its application in the prevention and treatment of AS was provided with theoretical guidance.HCAEC was treated with H2O2 to establish a cell damage model.PLA and HMGCR were moleculatively docked.Cy-totoxicity was detected by the CCK8 method,morphological changes of cells were observed by microscope,and TC content in HCAEC was determined by TC kit.The expression level of HMGCR in HCAEC was detected by immunofluorescence and Western blot.The results sho-wed that PLA has strong interaction with HMGCR,and the survival rate and morphology of cells were significantly improved,TC content in HCAEC and HMGCR expression level were decreased under the intervention of PLA.PLA can significantly inhibit the expression of HMGCR in HCAEC by interacting with HMGCR,thus reducing the level of intracellular TC,providing protection for Human coronary ar-tery endothelial cells injury,and providing theoretical basis for the prevention and treatment of AS.
