Study on the Extraction of Total Flavonoids From Pesudognaphalium affine(D.DON)Anderb.and Their Antioxidant Properties by Ultrasonic-assisted Two-phase Aqueous System
In this experiment,the best technological route of the extraction of total flavonoids by ultrasound-assisted aqueous two-phase from Pesudognaphalium affine(D.DON)Anderb.was explored by using Pesudognaphalium affine(D.DON)Anderb.as raw material and the yield of total flavonoids from Pesudognaphalium affine(D.DON)Anderb.as an index.Using 52×3 two-water phase test method,the optimum composition system of two-water phase was studied based on the yield of total flavonoids of Pesudognaphalium affine(D.DON)Anderb.The results showed that the optimal system composition was 16%dipotassium hydrogen phosphate and 30%ethanol.Compared with the traditional extraction methods of ethanol and hot water extraction,the two-aqueous phase method had better extraction effect,and the extraction rate was increased by 1 to 2 times.Taking the yield of Pesudognaphalium affine(D.DON)Anderb.as the response index,the optimal ultrasonic process parameters were optimized by response surface optimization,including ultrasonic power 420 W,extraction time 23 min,and ultrasonic temperature 50 ℃.Compared with the conventional aqueous two-phase extraction at room temperature,the extraction yield was increased by 34%.The results of the total antioxidant capacity test highlighted that the total antioxidant capacity of the flavonoid extract of the ultrasonic assisted aqueous phase was about 2 times that of the water extract and 1.8 times that of the ethanol extract.In con-clusion,the ultrasound-assisted aqueous two-phase extraction method optimized in this study has a higher extraction efficiency for the fla-vonoids of Pesudognaphalium affine(D.DON)Anderb.,and at the same time,the extract can maintain a stronger antioxidant activity.
