避瘟解毒颗粒对LPS诱导急性肺损伤小鼠保护作用的研究

探究避瘟解毒颗粒对脂多糖(LPS)致急性肺损伤(ALI)小鼠的保护作用。将30只C57小鼠随机分为正常组、模型组、地塞米松组、避瘟解毒颗粒高、低剂量组,每组6只,每天给药1次,连续给药7 d。模型组、地塞米松组、避瘟解毒颗粒给药组气管滴注脂多糖(LPS)构建小鼠急性肺损伤模型(ALI)。造模后72 h取材检测,分析小鼠体重变化情况;血常规指标;胸腺指数;ELISA法检测肺泡灌洗液(BALF)中IL-1β、IL-6、TNF-α水平;HE染色法观察肺组织病理变化。与模型组比较,避瘟解毒颗粒高剂量组小鼠血清中血常规指标BALF中炎性细胞因子IL-6、TNF-α含量,肺组织损伤程度均有不同程度的降低(P<0。05,P<0。01),且高剂量组能显著升高ALI小鼠的胸腺指数(P<0。01)。避瘟解毒颗粒对LPS诱导急性肺损伤发挥一定的保护作用,改善小鼠肺组织的病理改变,增强免疫功能,其作用机制可能与抑制促炎细胞因子的分泌及发挥抗炎作用有关。
Protective Effect of BiWen Jiedu Granules on LPS-induced Acute Lung Injury in Mice
To explore the effect BiWen Jiedu Granules on LPS-induced acute lung injury in mice.Thirty male C57 mice were randomly di-vided into the normal group,the model group,the dexamethasone group,and the low-dose and high-dose groups of BiWen Jiedu Granules,with 6 in each group,which were administered once a day for 7 days.The acute lung injury model was established by tracheal infusion of lipopolysaccharide(LPS)in the model group,the dexamethasone group,and the low-dose and high-dose groups of BiWen Jiedu Granules group.The material was taken at 72 h,the rats had their body weights,blood routine detected,their bronchoalveolar lavage fluid(BALF)lev-els of IL-1β,IL-6 and TNF-α detected by ELISA;the lung histopathological changes were observed by the HE staining.Compared with the modeld group,the the high-dose BiWen Jiedu Granules group shared lower degree of blood routine,levels of inflammatory cytokine IL-6 and TNF-αin BALF,lung injury(P<0.05,P<0.01),high-dose groups can increase the thymus index of ALI mice obviously(P<0.01).BiWen Jiedu Granules plays a protective role in LPS-induced acute lung injury in mice,improve the histopathological changes of lung,enhance immune function and its mechanism may be related to inhibiting the secretion of inflammatory cytokines function.
