Establishment of a Micro-Tissue DNA Extraction Method Suitable for the Identification of Sika Deer-Derived Products
In order to establish an efficient and convenient method for microtissue genomic DNA extraction suitable for molecular identi-fication of sika deer-derived products.In this study,fresh sika deer antler velvet,air-dried sika antler velvet,dried sika antler velvet,fresh sika deer whip,dried sika deer whip,air-dried sika deer whip,fresh sika venison meat,dried sika deer tail and sika deer blood were used as samples,genomic DNA was extracted by combining Chelex-100 resin solution with proteinase K,its concentration and purity were detected.Using this DNA as a template,the mitochondrial DNA D-loop region-specific gene of sika deer was amplified,the PCR amplification pro-ducts were electrophoresis and sequenced.The results showed that the concentration of genomic DNA template obtained by Chelex-100 re-sin extraction method was 52.57~63.39 ng/μL.A260/A280 ratio is 1.75~1.96.The electrophoresis band of mitochondria-specific gene am-plification products of sika deer is clear and the length is accurate;amplification product sequencing with similarity greater than 95%.Che-lex-100 resin extraction method can extract the genomic DNA of small amount of sika deer tissue samples and this method is cheap,simple and suitable for the authenticity identification of sika deer origin products.
