Salidroside Regulates the Biological Behavior of Acute Lymphoblastic Leu-kemia Cells by Inhibiting PI3K/AKT
To investigate whether salidroside regulate the biological behaviour of acute lymphoblastic leukaemia cells through inhibition of PI3K/AKT.This study stably cultured acute lymphoblastic leukaemia Jurkat cells were divided into control,SAL low dose,SAL medium dose and SAL high dose groups.MTT assay was used to determine the proliferation ability of cells in each group,flow cytometry was used to detect the apoptosis level,transwell assay was used to detect the invasion and migration ability of cells,and western blot was used to detect the expression level of P13K,AKT,and p-AKT proteins in cells in each group.MTT results showed that the cell growth inhibition rate of Jurkat cells in all groups increased with time,and the cell growth inhibition rate of Jurkat cells at all time points increased with the increase of SAL dose;flow cytometry results showed that SAL could increase the apoptotic rate of Jurkat cells,and with the increase of SAL concentration Jurkat cell apoptotic apoptosis rate gradually increased;transwell results showed that SAL could inhibit the invasive and migratory ability of Jurkat cells,and the invasive and migratory ability of Jurkat cells gradually decreased with the increase of SAL concentration;results of western blot assay showed that the expression levels of PI3K and p-AKT proteins were significantly down-regulated in all groups of cells after SAL treatment SAL is a novel therapeutic compound that inhibits the proliferation,migration and invasion of ALL cells and promotes the apoptosis of ALL cells.The anticancer effect of SAL may be achieved through the modulation of the PI3K-AKT signalling pathway.
