Analysis of Structure Characteristics of Promoter Region Sequence of TYR Gene in Chicken(Gallus gallus)
In order to analyze the structure characteristics of promoter region sequence of TYR gene in chicken,and the transcriptional regu-lation mechanism were preliminarily explored.Based on the 2 944 bp nucleotide sequence of 5'UTR and exon 1 of chicken TYR gene regis-tered in NCBI database,the core promoter region,basic regulatory elements,transcription factor binding sites and CpG island were analyzed by means of a variety of online softwares.The similarity of TYR gene promoter sequence between chicken and other species,at the same time,phylogenetic tree construction were carried out used DNASTAR.Lasergene 17.3 and MEGA 5.0.The results showed that the 1 500~2 100 bp upstream of initiaton codon of chicken TYR gene was a possible candidate core promoter region,and one potential transcription initiation site C was found at 1 604 bp of 5'UTR.There were one CAAT-box,nine TATA-boxes,and there was lack of CpG island in the promoter re-gion of TYR gene.The sequence also contains some transcription factor binding sites,such as C/EBPalp,GATA-1,Oct-1,TBP,Sp1,NF-1,API,SRF and NF-κB.The similarity of promoter region sequence for TYR gene between chicken and ring necked pheasant,turkey,quail,mallard,ruddy duck,crested duck,pig,common cow,water buffalo,hybrid cow,goat,sheep,red deer,white tailed deer,red deer and forest musk deer was 41.2%~89.7%.Phylogenetic tree revealed that chicken was closer with quail,ring necked pheasant,turkey and the farest re-lated was pig.This research results could provide theoretical basis for further revealing the transcriptional regulation mechanism of TYR gene for pigmentation trait of chicken.
