Curcumin Inhibits Proliferation and Induces Apoptosis in Papillary Thyroid Cancer Cells via PI3K/AKT Signalling
To study the effects of curcumin on PI3K/AKT signaling pathway in human thyroid papillary cancer cells(B-CPAP).The inhibi-tory effects of curcumin on B-CPAP cells at 12,24 and 48 h were detected by CCK-8 and clone formation assay.The apoptosis rate of B-CPAP cells was detected by TUNEL after 24 h treatment with different concentrations.Western blot analysis was performed to detect the expression of PI3K,p-AKT and AKT protein in B-CPAP cells treated with curcumin at different concentrations.The mRNA expression of caspase-3,BAX and BCL-2 in B-CPAP cells treated with curcumin at different concentrations was detected by RT-PCR.The CCK-8 assay showed that curcumin inhibited the proliferation of B-CPAP cells and increased with increasing doses.TUNEL method showed that B-CPAP cell apoptosis was significantly increased with curcumin doses increasing.After treatment of B-CPAP cells,the expression levels of PI3K,p-AKT and BCL-2 were decreased,while the expression levels of caspase-3 and BAX were increased.Curcumin may inhibit the proliferation and induce apoptosis of thyroid papillary carcinoma cells by down-regulating PI3K/AKT signaling pathway.
