芦根清肺颗粒制备工艺及TLC质量标准研究

本研究旨在优化和建立芦根清肺颗粒制备工艺与质量标准。本研究以提取物干浸膏得率为指标,通过单因素试验和正交试验法优选工艺参数;对处方中的芦根、桔梗、橘红、鱼腥草和枇杷叶进行薄层色谱法的定性鉴别研究。结果表明,试验最终确定的芦根清肺颗粒最佳制备工艺条件为所有药味分别加10倍和8倍量水,煎煮2次(第一次煎煮前需浸泡2h以上),每次1。5h,滤液浓缩至相对密度1。05~1。10(60 ℃),与适量的赋形剂糊精与蔗糖(质量比为1:1),制成溶解性和成型性良好的颗粒;薄层色谱法(TLC)结果显示,斑点清晰,专属性强,阴性无干扰。本品制备工艺稳定可靠。芦根、桔梗、橘红、鱼腥草和枇杷叶的薄层色谱无干扰,重复性好,专属性强。
Study on the Preparation Technology and TLC Quality Standards of Lug-en Qingfei Granules
The aim of this study was to optimize the preparation technolagy and establish the quality standards of Lugen Qingfei granules.The process parameters were optimized by single factor test and orthogonal test method using the yield of dry extract as indexes.The thin layer chromatography(TLC)was performed to identify Rhizoma Phragmitis,Radix Platycodonis,Citri Exocarpium Rubrum,Houttuyniae Herba,Eriobotryae Folium.The optimal preparation process of Lugen Qingfei granules was determined by adding 10 times and 8 times of water to all the medicinal materials,decocting twice(soaking for more than 2 hours before the first decoction),each time for 1.5 h.The ob-tained filtrate was concentrated to a relative density of 1.05~1.10(60 ℃)and mixed with an appropriate amount of excipient(the mass ration is)to produce granules with good dissolubility and formability.The results of thin layer chromatography(TLC)showed clear spots,high specificity,and there was no interference when compared with the negative.The preparation process of this product is stable and reliable.The thin layer chromatography of Rhizoma Phragmitis,Radix Platycodonis,Citri Exocarpium Rubrum,Houttuyniae Herba,Eriobotryae Folium showed no interference,with good repeatability and strong specificity.
