重组角蛋白酶降解羽毛后对小麦浸种催芽和盆栽的施用效果研究

为了合理利用禽类生产副产物角蛋白和减轻对环境污染,选用Bacillus licheniformis N5 基因在大肠杆菌中表达角蛋白酶后降解羽毛,制备降解液进行小麦浸种催芽、盆栽小麦根灌施用.结果表明,用氨基酸质量浓度为0.164 mg/mL的羽毛降解液浸种,小麦发芽势和发芽率最高分别为82.7%和 97.3%,相比对照分别增加 7.7%和 6.4%,施加氨基酸质量浓度为 1.030 mg/mL的羽毛降解液,小麦生长指标和根生长均为最佳,鲜质量与对照相比提高 9%,干质量与对照相比提高 4.8%,株高与对照相比提高 6.4%,根长与对照相比提高 82%.可见羽毛降解液能促进小麦发芽和生长,进一步可以开发为氨基酸肥料.
Study on the effect of feather degradation by recombinant keratinase on soaking seeds to accelerate germination and pot culture of wheat
In order to make rational use of keratin,a by-product of poultry production,and reduce environmental pollution,Bacillus licheniformis N5 gene was selected to express keratinase in Escherichia coli,and then feather was degraded,and degradation solution was prepared for wheat seed soaking to accelerate germination and root irrigation of potted wheat.The results showed that the highest germination potential and germination rate of wheat were 82.7%and 97.3%,respectively,which were increased by 7.7%and 6.4%compared with the control group.The application of feather degradation solution with amino acid concentration of 1.03mg/mL was the best condition for wheat growth index and root growth,with fresh weight increased by 9%and dry weight increased by 4.8 compared with the control group.It can be seen that feather degradation liquid can promote wheat germination and growth,and can be further developed into amino acid fertilizer.
